Objective:The experiment is according as the basic theory of TCM, to investigate G. Pentaphyllum's influence on the activity of Superoxide Dismutase(SOD),Glutathione peroxidase (GSH-PX) and Catalase (CAT) in the skin of hireless photoageing mice , wich result from ultraviolet irradiation, as well as the content of hydrogen peroxide (H202), Malondialdehyde(MDA)and hydroxyproline(HYP) in their skin, and to investigate G. Pentaphy Hum's effect on the structure of the hireless photoageing mice.Methods: Healthy KM haireless mice were divided into eight groups randomly and gavaged respectively, that is negative Control Group (A), Model Group (B), external Low-G. Pentaphyllum-Dosage Group (C), external Medium- G. Pentaphyllum -Dosage Group (D), external High-G. Pentaphyllum -Dosage Group (E), oral Low- G. Pentaphyllum -Dosage Group (F), oral Medium-G. Pentaphyllum - Dosage Group (G), oral High-G. Pentaphyllum -Dosage Group (H).The mice in all groups but group A were irradiated to simulate skin photoaging by long-term ultraviolet irradiation. At the same time, The mice those in C, D, and E groups were given G. Pentaphyllum externally and which in F, G, H groups through mouth. Staining skin with HE, Under 40 times light microscope, the structure of dermal was observed. While the changes of the content of H2O2, MDA, HYP, as well as the activity of SOD, GSH-PX and CAT in the skin were measured by biochemical test method in each group of the experiment. Furthermore the mtDNA3867 deletion can be detected by PCR.Results: In model group, the activity of SOD, GSH-PX and CAT in dermal were decreased , while the contents of the MDA, H2O2 were increased seriously (P <0.05) , on the contrary, the content of HYP was reduced. The skin biopsies appear photoaging look. Compared with the group B, the activity of CAT in group D and G were made a great progress (P<0.05) just as it of SOD in group C, D, G and H; as well as the activity of GSH-PX was increased whereas the content of H202 was decreased in group D, E, F and H; and simultaneously, the content of MDA was declined in all groups which given herb. The detection with PCR technic display that there were much more mtDNA3867 deletion in the skin of the mice in the model group than in the group A, D, E,G...
