Ganduqing buccal tablets (calling GDQ for short) were made in long-period clinical practice and pharmacological research. Based on the research of preparation, technology, quality specification and emergency toxicology of GDQ, to do the empirical study of the correlated pharmacodynamic action and mechanism of immunifaction. Objective:Observe the effects of GDQ on the viral pneumonia mouse infected by influenza virus FM1 in following aspects: anti-virus, anti-inflammatory, fever-relieving, analgesic effect and the mechanism of cytoimmunity, to supply theoretical and experimental proofs in further clinical research. Methods:1. Mouse were inoculated intranasally with mouse lung-adapted influenza virus strain (FM1), and were treat with GDQ in high dose, middle dose and low dose. Ribavirin and Lianhua Qingwen (a kind of TCM decoction) were used as positive control and blank drugs respectivly. To observe: (1) Dead-protection effect of mouses infected by FM1. (2) Lung index and inhibition ratio of lung index. (3) Pathomorphological variation of lung tissue; and pathomorphological difference among groups by semiquantitative analysis. (4) Influence in thymus and spleen index.2. Make febrile models on rats and rarebit by DNP and ETX respectively; Make swelling models of auricle and foot on mice and rats by Xylene and egg albumen respectively; Make pain models on mice by thermal stimuls and chemical stimulus respectively;3. ELISA method was used to detect the content of cytokine IFN-γ,IL-10 in serum on mice infected by influenza virus FM1; SABC immunohistochemical method was used to detect the expression of cytokine TNF-α,ICAM-1,NF-κB P65 in paraffin section of lung tissue and the variation of T lymphocyte subgroup in paraffin section of spleen on mice infected by influenza virus FM1.Result:1. Main results of drug efficacy(1) The death rate of mouse descended obviously, which had a certain dose-effectrelationship. There was significant difference between GDQ-high & middle-dosegroup and model group (p<0.001), so was GDQ-low-dose group and model group(p<0.05).
