Research The Detection Of SARS-CoV With SPR Biosystem

SARS-CoV was a nightmare to Beijing people which had caused great tragedy anddread at 2003 spring. It is urgent to devise a method for SARS-CoV detection. To dothis, the thesis was researched.SPR biosystems are a series of instruments researched and manufactured by CuiDa-fu group with surface plasmon resonance theory. In order to exactly detectSARS-CoV rapidly, two biochips were developed. They were used in reference SPRbiosystem to detect SARS-CoV antigen and antibody to SARS-CoV.Carboxymethyl dextran-modified SPR biochip was developed. How to decide thepH of dilution buffer was studied too. This chip had been used to immobilize or detectantigen, antibody, nucleic acid marked with biotin and receptor cell of EPO.Reference SPR biochip modified with dextran was developed. The method ofmodification was original creation. Two different areas of one chip could exit in onechannel. One was detection area could be used to immobilize protein; the other wasreference area which cann't bind protein. Reference chip is fit for detection in cellsubstrate and serum.SPR biosystem was used to detect SARS-CoV in this thesis firstly. Two kinds ofSARS detection chip were developed. One chip was used to detect antibody toSARS-CoV, the other was used to detect SARS-CoV antigen. Sample of SARS-CoVcould be detected by them directly. The sensitivity of the latter has reached to1.66×10~4PFU/ml which is close to the clinical need.The facture of two detection chip can be applied to manufacture other virusdetection chip. In order to save the developing time of new virus antigen detectionchip, monoclonal antibody was replaced by antiserum in research. The replacementwill improve abilities to face emerging virus infection and reduce the cost of detectionchip. The researches of biochip and virus detection help to use the reference SPRbiosystem in clinical diagnosis.
