Electric Study The Impact Of Memory-related Signaling Pathways In Vascular Dementia Rats

Objectives: Vascular dementia (VD) is a syndrome of acquired intelligence capacity damage which comes from mental disorder coming from various kinds of cerebrovascular diseases. It's mainly clinical manifestation is remembrance and cognition functional impairment, accompanied by the disorders of speech, movement, visual trabecular spaces and personality. Consequently, the focal point and breakthrough about the study of VD is to improve effectively study and remembrance function. Acupuncture treatment of VD possesses multichannel and multilevel feature. The topic inlet is signal passageway correlated to study and remembrance function, we observed pathological changes of hippocampus of VD and three signal passageways correlated to study and remembrance function to elucidate mechanism of electric acupuncture in preventing and curing VD. Methods: SD rats were randomly divided into 6 groups, namely the normal group, the sham operation group, the model group, the first electric acupuncture group, the second electric acupuncture group and the electric acupuncture pretreatment group. To adopt intraluminal thread approach of arteria cerebri media to replicate VD model of rats, we applied electric acupuncture to needle Baihui, Kidney Shu and Zusanli acupoint to treat VD, then detected study and remembrance function of rats applying Morries water maze, observed pathological changes of hippocampus microstructure of rats with HE dyeing and electron microscope, detected the content of nitric oxide (NO)and activity of induced nitric oxide synthase(iNOS) and content of glumatic acid of rats' hippocampus using radioimmunity method, observed the protein level of prekallikrein activator(PKA) and cAMP response element binding(CREB) of rats' hippocampus using immunohistochemical and western blot methods, detected N-methyl-D-aspartate receptor(NMDAR) mRNA expression of rats' hippocampus using in situ hybridization and observed the effects of above-mentioned indexes after electric acupuncture intervention.Results: (1)The escape incubative stage of rats in electric acupuncture group and electric acupuncture pretreatment group decurtated (P<0.01) and the result of fixing navigation experiment improved obviously (P<0.01), especially the curative effect of electric acupuncture one group was fine. (2)The pathological changes of rats' hippocampus of model group was significant, the electric acupuncture and the electric acupuncture pretreatment could relieve pathological damages. (3) Compared with that of the model group, the content of NO and activity of iNOS and content of cyclic guanylic acid of rats' hippocampus in electric acupuncture one group, the second electric acupuncture group, electric acupuncture pretreatment group decreased significantly (P<0.01 or 0.05). The difference of that among electric acupuncture one group, the second electric acupuncture group, the electric acupuncture pretreatment group wasn't significant in statistics(P>0.05). (4)Compared with that of the sham operation group, content of cyclic adenosine monomphsopate of rats' hippocampus in model group decreased significantly (P<0.01). Compared with that of the model group, content of cyclic adenosine monomphsopate of rats' hippocampus in the model group decreased significantly in electric acupuncture one group, the second electric acupuncture group, electric acupuncture pretreatment group heightened significantly (P<0.01 or 0.05). Compared with that of the sham operation group, the PKA and CREB protein expression of rats' hippocampus in model group decreased significantly (P<0.01); Compared with that of model group, the PKA and CREB protein expressions of rats' hippocampus in the first electric acupuncture group, the second electric acupuncture group, the electric acupuncture pretreatment group heightened significantly (P<0.01 or P<0.05). (5) Compared with that of the sham operation group, the glumatic acid content and NMDAR mRNA expression of the hippocampus of the rats in model group heightened significantly (P<0.01). Compared with that of model group, the glumatic acid content and NMDAR mRNA expression of rats' hippocampus in electric acupuncture one group, the second electric acupuncture group, electric acupuncture pretreatment group decreased significantly (P<0.01 or 0.05). The difference of that among electric acupuncture one group, the second electric acupuncture group, electric acupuncture pretreatment group wasn't significant in statistics(P>0.05).Conclusions: The electric acupuncture can improve effectively study and remembrance function of VD. It's mechanism of action comes true through some pathways as follows possiblely: The electric acupuncture can depress neurovirulence of NO and glumatic acid, regulate downward the content of cyclic guanylic acid, grow downwards necrosis of nerve cell, resist brain damaged through activating cAMP-PKA-CREB pathway, preserve brain cell, reduce apoptosis induced by glumatic acid -NMDAR pathway, lessen the destroy of long-term potentiation (LTP). The electric acupuncture can lessen nerve cell damage of cerebrovascular disease multichannelly, facilitate signal convery of study and remembrance function and promote amendment of study and rememberance function.
