Objective To study the antiangiogenic effects of Melittin on hepatocellular carcinoma and the relative mechanisms. Methods To study the inhibitory effects of Melittin on the proliferation of BEL-7402 cells and ECV304 cells by MTT assay, apoptosis and cell cycle distribution by flow cytometry, the migration and tubule formation of ECV304 cells by Transwell chamber assay and Matrigel assay, antiangiogenic effects by CAM test. To study the antitumor effects of Melittin on xenotransplanted models of nude mice. The concentration of VEGF, bFGF, and IL-8 was detected by ELISA. The mRNA expression of VEGF and bFGF was analyzed by RT-PCR. Expression of VEGF, bFGF, HIF-1αand NF-κB was deteced by immunohistochemical method and Western blot. Results Melittin could inhibit the proliferation of HCC cells and HUEVCs and angiogenesis in vitro.The result showed that Melittin could inhibit the growth and angiogenesis of transplanted tumor in dose-dependent manner in vivo. Conclusion Melittin has significant inhibitory effects on the HCC and the relative mechanism may be associated with the inhibition of angiogenesis.
