Objective:Hypertension is a common and frequently-occurring disease,which has a rising trend in the prevalence rate.The disabled and fatal complications caused by hypertension are often seen.Hypertension has become a major cardiovascular disease which threaten the health of the China's people seriously.With the thorough research on hypertension,it is found that there is a change in the vascular structure and function,which is called vascular remodeling.Vascular remodeling is not only the result of hemodynamic disorder and nervous-humoral change in hypertension,but also the important pathological basis which lead to the deterioration of hypertension and damage of target organs.The clinical Significance of vascular remodeling in hypertension has been paid extensive attention and become a hot point in research.At present,to reverse the vascular remodeling is one of the targeting in hypertension clinical treatment,which is the new studying field just springing up in world. Traditional acupuncture therapy has a definitely therapeutic effect on hypertension. Acupuncture has multi-facet,multi-level,multi-link adjusted role on blood pressure.Through the animal experiment,we could observe the effect of acupuncture on 2K1C-RHR carotid artery vascular remodeling,and detect the AngⅡ,ERK,Bcl-2,Bax,et al,in order to probe the possible mechanism of lowering the blood pressure and reversing the vascular remodeling using acupuncture,to find a new point of treating hypertension on acupuncture,and to provide a experiment basis for the studying of preventing and treating hypertension on acupuncture.Methods:SD rats were divided into five groups at random:the normal control group,sham operation group,model group,electroacupuncture treatment group, electroacupuncture treatment control group.To establish the 2K1C-RHR model, which was identified by measuring the rats' tail artery SBP,the model would be successful if the rats' tail artery SBP exceed 30mmHg more than the preoperative after 4 weeks,or the postoperative SBP is more than 20.0kPa(150mmHg). Electroacupuncture treatment group:we select the QuChi(pair);electroacupuncture treatment control group:we select the WaiGuan(pair).Using LH202H HanShi therapeutic equipment,selecting density wave,frequency 2Hz/15Hz,strength 1mA, stimulating 20 min every time,1 time a day,it is a couse of treatment in 8 weeks.We observe the change of the rats' carotid artery ultrastructure by electron microscope, detect the content of AngⅡin plasma and carotid artery by RIA,detect the protein expression of ACE,Bcl-2,Bax in carotid artery by immunohistochemical method, detect the apoptosis of VEMC in carotid artery by TUNEL,and detect the content of p-ERK by Western blotting.All the datum were processed by SPSS12.0 statistics software package and all results were demonstrated by mean±standard deviation.Results:(1)Compared with the normal control group,the systolic blood pressure of 2K1C-RHR elevated significantly(P<0.05),the vascular smooth muscle cells became hypertrophical,fiber node based organizations were of hyperplasia, mitochondria in the vascular smooth muscle cells was swelling,rough endoplasmic reticulum increased,vascular wall thickness(WT)increased,vascular diameter(LD) reduced,ratio of WT/LD enlarged,the vascular remodeling is obviously.After 8-weeks treatment,WT,LD,WT/LD and area of VSMCs improved in the electroacupuncture treatment group and electroacupuncture treatment control group (P<0.05).Through electron microscope,the structure of VSMCs were as same as the normal,cells were smaller than the model group,the figure of nuclear were normal,the increase of fiber outside cells were not obvious,mitochondria in the VSMCs was't obviously swelling,rough endoplasmic reticulum did't increased distinctly.(2)Compared with the normal control group,the content of AngⅡin plasma and carotid artery in the model group were both significantly increasing.After 8-weeks treatment,the content of AngⅡin plasma and carotid artery in the electroacupuncture treatment group and electroacupuncture treatment control group cut down obviously.There was a significant difference compared with the model group.Compared with the normal control group,the expression of ACE in carotid artery in the model group strengthened obviously.After 8-weeks treatment,the expression of ACE in carotid artery in the electroacupuncture treatment group and electroacupuncture treatment control group weakened,and there was a significant difference compared with the model group.(3)Compared with the normal control group,the apoptosis of VSMCs in Carotid artery of the model group increased,the expression of Bcl-2 increased and the expression of Bax decreased.After 8-weeks treatment,the apoptosis of VSMCs in carotid artery increased in the electroacupuncture treatment group,the expression of Bcl-2 decreased and the expression of Bax increased.There was't a significant difference in the apoptosis of VEMCs in carotid artery of the electroacupuncture treatment control group compared with the model group.(4)Compared with the normal control group,the expression of p-ERK in carotid artery increased in the model group.After 8-weeks treatment,the expression of p-ERK in carotid artery decreased in the electroacupuncture treatment group and electroacupuncture treatment control group,and the activity of ERK decreased.Conclusion:Electroacupuncture "QuChi" could decreases the systolic blood pressure and improves the pathological changes and vascular remolding in carotid artery of 2K1C-RHR significantly.The possible mechanism is:(1)Activating the ERK signal way,restraining the release of AngⅡ,improving the pathologicl change of VSMCs' hyperplasia caused by long-term hypertension and nervous endocrine excessive activation.(2)Activating the ERK signal way,restraining the unnormal apoptosis of VSMCs in pathological process of hypertension.The expression of Bcl-2 decreased and the expression of Bax increased.(3)The balance between apoptosis and proliferation was improved.The vascular remodeling in hypertension was alleviated and the pathological state in hypertension was mitigated. |
