Objective The nasopharyngeal carcinoma is one of the most common malignant tumors and threatening the people's health seriously.This project trits to establish a set of economic,fast,effective the experimental model of screening anti-NPC Chinese herbal medicine in vitro,in vivo and in serum pharmacology with the cell and molecular biology and so on modern experimental techniques and the NPC cell lines CNE1,CNE2,TWO3 and C666-1.we had screened the eight kinds of Chinese herbal medicine, including six kinds of simple drugs,Sarcandra,astragali radix,coicis semen,Stem or Leaf of Fortune Euonymus,Yetbadetajo Hert and three kinds of complex prescription,Biyan Qingdu granula(BYQDKL),Fufang Tianxian capsule(FFTXJN)and Yangyin Qingre Jiedu agent(YYQRJDJ)with this model, which providing a theoretical basis for the development and utilization of Chinese herbal medicine resources and the prevention and treatment of NPC.Research contents1.Establishing elementary model from screening Chinese herbal medicine to cure nasopharyngeal carcinoma(NPC)in vitro and elementary application Method We used the technique of the cell biology and the molecular biology such as MTT method,flow cytometer(FCM)method to establish the elementary model from screening Chinese herbal medicine to cure nasopharyngeal carcinoma(NPC)in vitro.Through the model,we had screened the eight kinds of Chinese herbal medicine.Result In MTr method,coicis semen,BYQDKL and FFTXJN could inhibit the growth of NPC cells and the inhibition was dose-dependent and time-dependent in 5 days.And the IC50is smaller than 50mg/L in 48h. Sarcandra,astragali radix,Stem or Leaf of Fortune Euonymus,Yetbadetajo Hert,YYQRJDJ could not inhibit the growth of NPC cells.In FCM method, coicis semen,BYQDKL and FFTXJN have strong function in promoting apoptosis to cells.but Sarcandra,astragali radix,Stem or Leaf of Fortune Euonymus,Yetbadetajo Hert,YYQRJDJ could not.The results were the same as the MTT method's.2.Establishing elementary model from screening Chinese herbal medicine to cure nasopharyngeal carcinoma(NPC)in serum pharmacology and elementary applicationMethod We used the technique of the cell biology and the molecular biology such as MTT method,flow cytometer(FCM)method to establish the elementary model from screening Chinese herbal medicine to cure nasopharyngeal carcinoma(NPC)in serum pharmacology.Through the model, we had screened the eight kinds of Chinese herbal medicine.Result In MTT method,Sarcandra,coicis semen,BYQDKL and FFTXJN could inhibit the growth of NPC cells and the inhibition was dose-dependent and time-dependent in 5 days and are bigger than 30%in 48h.astragali radix,Stem or Leaf of Fortune Euonymus,Yetbadetajo Hert,YYQRJDJ could not inhibit the growth of NPC cells.In FCM method,Sarcandra,coicis semen,BYQDKL and FFTXJN could make the NPC cells more in the period of G0/G1 and less in the period of G2/M compared with the control.3.Establishing elementary model from screening Chinese herbal medicine to cure nasopharyngeal carcinoma(NPC)in vivo and elementary applicationMethod The models of xenografted tumor in nude mice were established with CNE1 and CNE2.Morphology of xenografts were investigated by HE staining.Ultra micro-structured changes of Carcinoma cells were observed by transmission electron microscope(TEM).In TUNEL of histochemistry were used to detect the apoptosis of Carcinoma cells.Immunohistochemical method were used to detect the expression of Bcl-2-,Bax and VEGF.The telomerase activity was observed by using TRAP-ELISA method.Result Through the BALB/C-nu/nu mouse model,we can screen that Sarcandra and FFTXJN could significantly inhibit the growth of tumor when compared with the control group.The number of tumor cells decreased gradually with Sarcandra groups,FFTXJN groups,coicis semen groups and the number of fibroblasts increased.Electron microscopy indicated the apoptosis of tumor cells with marginal nuclei,chromatin condensation and nuclei fragmentation,and apoptotic bodies were observed.The rate of cell apoptosis for this three groups were higher than that of the control.The expression of Bcl-2,VEGF were lower and the expression of Bax of the three were higher than that of the control.The telomerase activity were lower than that of the control.Conclusion We have established a set of economy,fast,effective the elementary model in vitro,in vivo and in serum pharmacology.Through the model,we screened that Sarcandra and FFTXJN were effective anti-drug for NPC.
