| Objective:To investigate effects of Traditional Chinese Medicine Yitangkang on protein content and gene expression of Glucose Transporter 4(GLUT4) in skeletal muscles of Type 2 diabetic rats.And to thorough research the underlying molecular mechanism of Yitangkang causing transduction of GLUT4.Method: Type 2 diabetic rat models were established by feeding with high-lipid diet and intraperitoneal injection of small-dose streptozotocin (STZ) once.After 4 weeks treating by Yitangkang,rats were examined fasting blood glucose and fasting serum insulin.Then,killed rats , took skeletal muscle,and detected the change of expression of GLUT4's protein and mRNA. At the same time,by western blot method , checked the expression of protein kinase B and phosphatidylinositol 3-kinase;Using reverse transcription-polymerase chain reaction to measure adapter protein.Oncogene protein mRNA expressive level,compared with model group,to conjecture and judge the exact mechanism of Yitangkang to cause GLUT4 transfer.Results:Compared with normal group,fasting blood glucose,blood fat and fasting serum insulin of Type 2 diabetic rats increased obviously(P<0.01).While Insulin sensitive indication(ISI)decreased(P<0.01),GLUT4's protein and mRNA expressive content both went down(P<0.05);Compared with model group,treating group′s fasting blood glucose decreased obviously(P<0.01),serum insulin went down(P<0.05),ISI went up(P<0.01),GLUT4's protein and mRNA expressive content increased (P<0.05),but still lower than normal group.Furthermore,Yitangkang could increase the statements of Protein kinase B and phosphatidylinositol 3-kinase in skeletal muscles,and yet have no function on gene expression of adopter protein and oncogene protein.Conclusion : Feeding with high-lipid diet and intraperitoneal injection of small-dose streptozotocin (STZ) can establish an animal pattern of Type 2 diabetic rats. Traditional Chinese Medicine Yitangkang can not only improve the abnormal glucose load but also improve insulin resistance. This mechanism mainly rely on increasing the statements of Protein kinase B and phosphatidylinositol 3-kinase in skeletal muscles,which could ameliorate GLUT4 transfer mechanism and increase GLUT4's protein and mRNA expression.The function is relative to increasing action of skeletal muscle absorbing and making use of glucose . |
