Objective: Ruoshiming Oral Liquid is TCM new drug for teen-ager amblyopic therapy. In order to study the drug' function of improving visual electro-physiology, protecting mice's retina light injury, promoting blood flow, protecting ganglion cells, eliminating free radical& protecting vascular function, et al, we modelled amblyopia and retina injury anmal, and then evaluated the drug's main pharmacodynamics and mechanism.Methods: Suture methods was used to establish deprived amblyopic rat models and then we observed the flash visual evoked potential changes with visual electro-physiology instrument. The retinal photic injury rat models were established by light damage trunk, and we observed the number and mensurated the nucleolus content of the retinal outer nuclear layer nerve cells' pathological changes with optical microscope. The acute blood-stasis rat models were estabilshed by injecting adrenaline, and we detected the whole blood viscosity in the rats with auto-viscometer. To study the mechanism, suture methods was used to establish deprived amblyopic rat models and we observed the number of retina ganglion cells, the size of nucleolus & pathomorphism change with optical microscope. Lipid peroxidation (LPO) was measured with thiobarbituric acid chromatometry and the method of pyrograllol autoxidation was used to determine superoxide dismutase (SOD) . We measured the eyes' concentration of nitric oxide (NO) and the activity of nitric oxide synthase (NOS) by the corresponding kits to illuminate the mechanism of Ruoshiming Oral Liquid action. All data were processed by SPSS13. 0 for Windows and denoted with (x|-)±s.Results: The pharmacodynamics studies show that Ruoshiming Oral Liquid could reduce the amblyopic eyes' elongation of FVEP N1 trough & P1 wave crests' latent period and enlarge the amblyopic eyes' FVEP N1-P1 amplitude. The drug also could increase retinal outer nuclear layer cells and protect nucleus of retinal nerve cell and promote blood circulation.It has been discovered in mechanism researches that Ruoshiming Oral Liquid could increase retinal ganglion cell number, improve pathological changes in nucleus of ganglion cells, alleviate atrophy of nucleus, improve pathological changes in ganglion cells. It could also increase the activities of SOD & NOS, improve the NO content and decrease LPO level, prevent oxygen free radical injury, protect vascular endothelium and improve vascular function.Conclusion: It has been discovered in experimental researches that Ruoshiming Oral Liquid acts on therapy of amblyopia. Its mechanism may be associated with protection of retinal ganglion cells and elimination free radical, dilation of blood vessel, protection vascular endothelium, improvement vascular function.
