Chronic obstructive pulmonary disease(COPD) is a important cause of growing morbidity and mortality in worldwide respiratory system It is characterized by incompletely reversible limited gas flow and gradually decreased lung function. The pathogenesy of COPD has not been fully understood yet. We now generally consider that COPD is a kind of chronic inflammation in its airway and lung parenchyma and lung blood vessel. COPD has increased alveolar macrophage and T lymphocyte,especially CD+8,and neutrophil in the different places of lung. Activated inflammatory cells can release many kinds of mediators. These mediators can destroy lung construction and promote inflammation reaction of neutrophil. Besides inflammation, the imbalance of lung protease and antiprotease and of oxidation and antioxidation also has an important effect in the development of COPD. Its occurrence and development is intimately correlated with various tendency factor, cell, growth factor and enzyme. In our study, we used immunohistochimstry and semi-quantitative reverse transcription-polymerase chain reaction(RT-PCR) and western blotting to detect the expression of cyclooxygenase-2(COX-2) and platelet-derived growth factor A(PDGF-A) and platelet-derived growth factor B(PDGF-B) in the lung tissues and its alveolar epithelial cell. We also observed the effect of the cytokine of interleukin 4(IL-4) in our experiment. Finally we investigate the nosogenesis and prevention and cure of COPD. Twenty-four rats were randomly divided into the control group, the model group and the dexamethasone group. Every group had eight rats. The expressions of COX-2,PDGF-A and PDGF-B in the lung tissue were detected by immunhistochemistry, semi-quantitative RT-PCR and western blotting.Results Compared with the control group,the expressions of COX-2,PDGF-A and PDGF-B in the model group and dexamethasone group were increased significantly. And compared with the model group, those expressions were decreased obviously in the dexamethasone group.Conclusion The expressions of COX-2 and PDGF in the lung tissue of COPD were increased significantly and dexamethasone can decrease those expressions.
