Objective To investigat the differentiation of human adipose derived stem cells (hADSCs) into cells of retinal pigment epithelial(RPE),photoreceptor and vascular endothelial in vitro and in vivo.and study the hADSCs' effects on rats' retinal degeneration induced by NaIO3.Methods hADSCs were isolated,cultured and identified,hADSCs were induced into cells expressing endothelium-specific marker vWF,RPE-specific marker Pan-CK and photoreceptor-specific marker rhodopsin in vitro.Rats retinitis degeneration models were established.60 rats were randomly distributed into 6 groups.In each group,5 rats were injected with hADSCs,while the other 5 rats were injected with PBS through tail vein. The rats were sacrificed and the eyes were enucleated by surgery separately.Paraffin wax sections were stained with anti-PCNA,anti-human cell nucleus, anti-rhodopsin,anti-MITF or anti-vWF monoclonal antibody respectively to analyze the ADSCs' differentiation and the effects on treatment of retinal degenerationResults hADSCs could be induced to express endothelium-specific marker, RPE-specific marker and photoreceptor-specific marker in vitro.The reparation and proliferation of retina in the hADSCs-transplanted groups were better than those in the control groups.By 1 and 2 weeks after transplantation,hADSCs engrafted into the host retina and choriocapillary.By 3 weeks,transplanted cells expressed the RPE-specific marker MITF,endothelium-specific marker vWF and photoreceptor-specific marker rhodopsin..Conclusions hADSCs have the ability to be induced into cells expressing RPE-specific,endothelium-specific and photoreceptor-specific markers in vitro and in vivo.hADSCs can promote the reparation and proliferation of damaged retina.
