All kinds of biologic characteristic of HCC reflect its malignant degree.Modern multi-modality molecular imaging fusion technique,such as PET/CT,provides a new method to evaluate HCC.The study comes from the insufficiency problem of HCC's biologic characteristic imaging.The aim is to image the HCC's different biologic characteristic using multiple different positron molecular probes as a combination mode.So we conducted the following research.Four different biologic characteristic HCC cells line,including high and low oncogenicity cell line QGY7701 and HEP G2,and high and low metastatic cells line MHCC-97H and MHCC-97L cells line,were brought into this research field.And the lung carcinoma cell lines(A549) was as a positive contrast group.Cells were grown as a monolayer in DMEM medium.All of Cells were performanced positron tracers blinding experiment.The tracers are FDG and FLT and CH.Then we may draw a pattern of molecular probes combination.In addition,four different biologic characteristic HCC cells line,were performanced tracer blind experiment under the different intervention methods.The intervention methods include increasing glucose or insulin level in DMEM medium,and the chemotherapeutics 5-FU 1ug/ml with DMEM medium.The Radionuclide uptake and efflux assay were also conducted. According to the intervention experiment results,we may optimize the pattern of molecular probes combination.Ultimately,we can establish a lookup table of diagnosis and a discriminational squence to classify the biologic characteristic of HCC cell lines.Some molecular biologic indexs which were relation to tracers' uptake were analyzed,and comparing with cells uptake tracer's results.These biologic factors include OPN,Glut1,Glut3 and Ki-67 in fours different HCC cells lines expression.Animal models who burden different biologic characteristic HCC were performanced imaging by mircoPET/CT system.A single probe of three tracers was no different between different metastatic cells. However,there was a diiferent of FDG and FLT uptake ratio between different oncogenicity cells.and but there was no different of CH uptake ratio between different oncogenicity cells.There were different in outflow and absorb ratio of four HCC cell lines uptake tracers.There was a positive correlation between outflow and absorb ratio.The outflow and absorb ratio of FLT were higher than FDG.Under high glucose levels intervene,the FDG uptake ratios of cells were decrease.Under high insulin level intervenes,the FDG uptake ratio of cells were increase.There are no different between the cells uptake ratio of 5-FU chemotherapeutics with 24h intervention.In the microPET/CT imaging part,the FDG T/NT of different oncogenicity mice models was higher than of different metastasis.The QGY7701 mice model was the most highest in uptake FDG and FLT.Mice model imaging results were the same completely as cells line experiment results.There were different among OPN,Glutl, Glut3,ki-67 expressions in four cells line.And biologic factors expression index were higher in the different metastatic cells line than the different oncogenicity cells line. There was no relation with the different metastatic cells line between uptake FDG and OPN,Glut1,Glut3,ki-67.There was a positive correlation with the FDG uptake ratio of different oncogenicity cells line.The OPN,ki-67 with FLT uptake ratio was a positive correlation,but was a negative correlation with different oncogenicity cells line,and no correlation with CH uptake ratio.The OPN,Glut1,Glut3,ki-67 expressions indexes in animal tumor result were same as cell experiment results and correlation with microPET/CT imaging results.According to the results of statistic analysis,we established a lookup table of diagnosis to classify the different biological characteristic of HCC.The lookup table of diagnosis and the discriminational sequence can classify the biological characteristic of HCC.The animal experiment may reflect the cell experiment result with discrimination sequence and lookup table of diagnosis.
