Objective At present, the characteristic of younger age, urbanization and expansion have been initially shaped in breast cancer. Invasive breast cancer is the most common type of breast carcinomas. To study the change and express of relative gene by advanced molecular biology technology is the new field in the human cancer research, and the effect of environment on the breast cancer has been paid more attention. Our aim is to explore the living environmental factor which has impact on the breast cancer, and to provide scientific basis for making prevention measure to it. Meanwhile, our purpose is to find new molecular biologic therapeutic target for breast cancer by studying the express and imprinting status of IGF2 in invasive breast cancer.Objects and methods1. Case-control was used in this study. 121 cases were included in this epidemiological study; all of them were diagnosed as invasive breast carcinomas by pathological diagnosis in Hubei Province Cancer Hospital and Wuhan Central Hospital from March to July, 2007, habitation-matched 127 health women beyond 35 years old were as controls.2. 63 pathological samples of invasive breast cancer and matched breast tissues five centimeter away from the tumors and 70 breast benign lesions which have been diagnosed in the same time in the above two hospitals were included in this part study. We analyzed the express difference of IGF2 in the three groups and the living environmental factors which had effect on IGF2 status.3. This part study included 47 invasive breast cancer tissues and matched breast tissues five centimeter away from the tumors; we detected the imprinting status of IGF2 in the tumor tissues and adjacent breast tissues.4. Single factors analysis was performed by t andχ2 test, and Logistic model was used to analyze the impctor of the breast cancer.Results1. The sort of these factors which have positive effect on the risk of breast cancer is stimulated by negative incidents with 5 years, usually dyed fair and excited by noise, weight on meats in their diets according to the contribution to the breast cancer of these factors. But the order of protective factors to the breast carcinoma is to eat spicy foods sometimes, drinking milks and preparation and drinking tea often.In pre-menopausal women, often eating animal meats, dyed fair, stimulated by noise and the feeling of loneliness have positive relation to the risk of breast cancer, but eating astacus and fishes often, consume the marine products and abreaction at the right moment are the protective factors to the carcinogenesis of breast. For postmenopausal, drinking milks and preparation often, mild in salt consumption, eating spicy foods and acids foods sometimes have negative effect on the risk of breast cancer.2. The express incidence of IGF2 in invasive breast cancer tissues is much higher than that of in breast benign and adjacent breast tissues, the difference has statistical significance(χ2=13.35,4.37;P =0.0003,0.0365), but there is no statistical difference in express incidence between the breast benign and adjacent breast tissues(χ2=3.26,P =0.0709). The express of IGF2 in invasive breast cancer tissues has positive relation to the histological grade of tumors, transfer of lymphoid node and status of ER. Drinking tea often and increased BMI are the dangerous factors to the positive express of IGF2.3. 24 Heterozygote samples were detected in 47 breast cancer tissues. Among these heterozygote samples, 12 samples had LOI of IGF2 in breast cancer tissues and 19 samples had LOI of IGF2 in adjacent breast tissues, the difference between the groups has statistical significance (P =0.035).Conclusion1. Living environment has influence on the risk of breast carcinomas, preventive measures should be made according to the roles of these factors, and these measures can decrease the risk of breast cancer.2. IGF2 is involved in the process of breast cancer, and environmental factors have effect on the express of IGF2 in breast tissues. But the increase of IGF2 express in breast cancer tissues is not because of LOI.InnovationWe combined the micro with macro study in this study, considering the environmental and gene factors which have effect on the risk of breast cancer and the molecular biological parameter and environmental factors were synthetic approached.
