Objective: To probe the pathological foundation andessence of Yin deficiency syndrome in TCM from two anglesof protein molecule and gene by investigating the levelsof interleukin-1β(IL-1β),interleukin-6 (IL-6) andtumour necrosis factor-α(TNF-α) of the patientsdiagnosed as Yin deficiency syndrome in perimenopausalsyndrome.Methods: Select 30 cases of perimenopausal syndrome bothdiagnosed as Yin deficiency syndrome which are used asperimenopausal syndrome -Yin deficiency group, and 30healthy persons are used as control group. The bloodsampling from vein of all cases of both groups wasperformed before meal in early morning. After the patientsof perimenopausal syndrome-Yin deficiency group takeHerbal Preparation for Yin Deficiency Syndrome 2 months,we performed the blood sampling at the same conditions.(1) Fasting serum levels of IL-1β,IL-6,TNF-αin allcases of the both groups were determined by anenzyme-linked immunoadsordent assay (ELISA) method withdouble antibody and sandwich technique.(2) IL-1β,IL-6,TNF-αmRNA expressions in all cases ofthese groups were observed by reverse transcriptase-polymerase chain reaction (RT-PCR).Results: (1) The levels of serum IL-1β,IL-6,TNF-αin the cases of both perimenopausal syndrome- Yin deficiency group after 2 months treatment by nourishingYin preparation and control group were significantlylower than those in perimenopausal syndrome- Yindeficiency group before treatment (P<0.01), however,there is no significant difference in IL-1β,IL-6,TNF-αlevel between the control group and perimenopausalsyndrome- Yin deficiency group after 2 months treatment.(2) The expressions of IL-1β,IL-6,TNF-αmRNA in thecases of both perimenopausal syndrome- Yin deficiencygroup after 2 months treatment and control group weresignificantly lower than those in perimenopausalsyndrome- Yin deficiency group before treatment(P<0.01),however, there is no significant difference in IL-1β,IL-6,TNF-αlevel between the control group andperimenopausal syndrome- Yin deficiency group after 2months treatment.Conclusions: perimenopausal syndrome patients are in astate of immune activation. The difference of serum IL-1β,IL-6,TNF-αlevel between before and after treatmentof nourishing Yin herbal preparation may be a sensitiveparameter of illness recovery.IL-1β,IL-6,TNF-αlevel in the patients sufferingfrom perimenopausal syndrome are possibly related closelywith Yin deficiency syndrome.Since the expression of IL-1β,IL-6,TNF-αmRNAinfluences the serum IL-1β,IL-6,TNF-αlevel, theheightened expression of IL-1β,IL-6,TNF-αmRNA may be associated with occurrence of Yin deficiency syndrome inTCM.
