Objective:(1)Through the literature review, explain the pathogenesis of Postmenopausal women with coronary heart disease.and the similarities and differences with Internal medicine coronary heart disease. To dicuss the different recognize and treatment of Postmenopausal women with coronary heart disease between Chinese medicine and morden medicine.(2)Observed the change of ovariectomized female rabbit serum sex hormones level, lipids level, etc. after they are feed on ordinary diet and high fat diet, research the change of sex hormones and blood lipid in ovariectomized rabbit,and the relevant between sex hormones and serum lipids.and the impact of these factor in the high fat diet feeding.(3)Observe the change of serum sex hormones, lipids, endothelial cell apoptosis, smooth muscle cell proliferation and so on,after Zishenningxin recipe and Estrogen Replacement Therapy impact on ovariectomized female rabbit with high fat diet. from the regulation of blood lipids, improvement of endothelial apoptosis, inhibition of smooth muscle hyperplasia, etc. explore the mechanisms of Zishenningxin recipe prevent and treat the heart of coronary heart disease in postmenopausal.Methods:(1)Collection of literature, explore the pathogenesis and treatment methods of postmenopausal coronary heart disease from Chinese medicine and Western medicine.(2)27 ovariectomized female rabbits were randomly divided into sham-operated group, OVX group, OVX high-fat group every groups is 9. feeding the corresponding feed for 12 weeks. The end of the experiment, after fasting for 12 hours, cardiac blood serum samples collected, TG, TC, HDL-C were determinate by enzymatic method; E2, FSH, LH using radioimmunoassay method. killed animals by Air embolization, aortic and uterine using HE staining to observe morphological changes;(3)36 ovariectomized female rabbits were randomly divided into sham-operated group, model group, zishenningxin group and Premarin group, every groups is 9. the last three groups feeding the high fat diet, Zishenningxin recipe Group and Premarin group gavage 1 time daily for 12 weeks. The end of the experiment, after fasting for 12 hours, cardiac blood serum samples collected, TG, TC, HDL-C were detected by enzymatic method; E2, FSH, LH were detected using radioimmunoassay method. killed animals by Air embolization,using HE staining to observe morphological changes of aortic, the expression change of ER, Bcl-2, Bax, PCNA were detected by immunohistochemistry method, the expression change of PDGF-BmRNA, VCAM-1mRNA were detected by RT-PCR, the endothelial cell apoptosis rate detected by Flow cytometry assay.Results:(1) Chinese medicine think the root causes of coronary heart disease in postmenopausal are Kidney deficiency,the main treatment of coronary heart disease in postmenopausal are based on Kidney-tonifying, morden medicine think mainly because of the estrogen reducing the protective effect to cardiovascular, so the sex hormones replacement therapy is the main treasure, but its role is still in dispute.(2) E2 level among the three groups has no significant difference. Among the three groups FSH, LH levels were significantly different(P<0.05), OVX group, OVX high-fat group compare to sham-operated group were significantly increased, the results have statistical significance (P<0.05). OVX high-fat group TC, LDL, HDL levels and sham-operated group, OVX group has significant difference (P<0.05), TG levels among the three groups showed no statistical significance. sham-operated group and OVX group intimal smooth, no plaque formation; OVX high-fat group endometrium has a white resin-like material protruding to the lumen; HE staining can be seen that OVX group membrane there is a slight thickening, OVX high-fat group significantly intimal thickening, plaque obvious bulge, containing a large number of foam cells.(3)①E2 levels among the four groups has no significant difference; FSH, LH levels, model group, Premarin group compare to sham-operated group has significant difference, zishenningxin group compare to sham-operated group has no significant difference, LH level of sham-operated group and model group has significantly different(P<0.05).②TC, LDL-C levels, model group, zishenningxin group, Premarin group compare to sham-operated group has significant difference(P<0.05), zishenningxin group, Premarin group compare to model group have significant difference(P<0.05),there is no difference between the two drugs; TG levels, there is no significant difference among the four groups; HDL-C levels, model group, zishenningxin group, Premarin group compare to sham-operated group has significantly difference, zishenningxin group, Premarin group compare to model group were significantly different, but the two drugs lower the HDL-C level, APOA1 levels, zishenningxin group, Premarin group compare to model group have significant difference. (3) The expression of estrogen receptor of each group, sham-operated group was significantly higher than other groups, model group, Premarin group compare to sham-operated group, the results have statistical difference (P<0.05), zishenningxin group compare to sham-operated group was not significant. zishenningxin group, Premarin group compare to model group, the expression increases in both, but in zishenningxin group increased more significantly.④model group significantly intimal thickening, plaque obvious bulge, containing a large number of foam cells. The lesions of zishenningxin group, Premarin group have alleviated in varying degrees than model group.⑤endothelial cell apoptosis:there is no statistical difference between each group, model group, zishenningxin group, Premarin group compare to sham-operated group have both increased apoptosis rate, zishenningxin group, Premarin group compare to model group the apoptosis rate decrease. Bcl-2 expression, model group,zishenningxin group, Premarin group compare to sham-operated group has decreased, zishenningxin group, Premarin group compare to model group have increased; Bax expression, model group, Premarin group compare to sham-operated group has increased, zishenningxin group, Premarin group compare to model group has a larger lower. there is no significant difference between two groups.⑥proliferation of smooth:model group compare to sham-operated group (P<0.05), PCNA expression shows castration add to high-fat can promote the role of smooth muscle cell proliferation, zishenningxin group, Premarin group compare to model group differences were significant (P<0.05), but there is no significant difference between the two drugs. PDGF-B mRNA expression in each group were significant difference compare to sham-operated group, (P<0.05), zishenningxin group, Premarin group compare to model group there is significantly difference (P <0.05), but there is no statistics difference between the two drugs.⑦of adhesion molecule-1 (VCAM-1):VCAM-1mRNA expression in each group compare to sham-operated group, there is significantly difference (P<0.05), zishenningxin group and Premarin group compare to model group has significant (P<0.05), but is no statistics difference between the two drugs.Conclusion:①Zibushenyin is the mainly treasure to postmenopausal coronary heart disease, morden medicine think HRT is the maily treasure.②ovariectomized female rabbits can not form typical AS model with normal feed.but high fat feed can.③Zishenningxin recipe can regulate hormone levels, elevated E2 levels, lower FSH, LH level, corrective endocrine disorders; lowering LDL-C, TC, improve blood lipid metabolism, atherosclerosis confrontation; increased estrogen receptor vitality of aortic, and inhibit vascular endothelial cell apoptosis, the protection of vascular endothelial integrity, inhibit smooth muscle hyperplasia, to affect some aspects of the occurrence of coronary heart disease in postmenopausal development.
