AIMRunx2 (Runt-related gene 2,Runx2) is a member of the Runx family of transcription factors with a restricted expression pattern. It has so far been detected predominantly in skeletal tissues where, inter alia, It regulates osteoblast diferentiation,development and bone formation. Galectin-3 belongs to the galectin family of P-galactoside-binding soluble lectins, it has been implicated in cell and cell,celland cell-matrix interactions.there is increasing evidence show that for a role of Runx2 and Galectin-3 in immune response and infmation as well as in normal developmental processes, involved in cell growth and differentiation,apoptosis regulation,cancer prognosis,cytokine production, etc. they were expressed in many tumors, There was little literature about Runx2 and Galectin-3 expression in Central Nervous System.our study aimd to explore the expression and relationship of Runx2 and Galectin-3 in glioma tumour, to elucidate the relationship between clinicopathological factors of Runx2 and Galectin-3.In Vitro study,we knock down the Runx2/Galectin-3 express by Small RNA interference technique and observe its effection on mRNA,protein level and cell proliferation activity in U251 cell. This study is to provide experimental and theoretical data in further investigation aimed on the target of Runx2 and Galectin-3 for glioma therapy.Methods1.50 surgieal specimens were obtained from Patients with gliomas of zhongnan hospital,wuhan university,including 26 males and 24 females, ranging in age from 10 to 65 years(mean41.7).According to world health organization,There were 11 gradeâ… ,8 gradeâ…¡,15 gradeâ…¢,16 gradeâ…£.The expression of Runx2 and Galectin-3 were detected by S-P immunityhistochemical method in 50 cases of human cerebral gliomas and Takes 10 example of normal brain organization make the comparison.2.In Vitro study,we select the valid siRNA and transfect the U251 cell,then incubate for 24h,48h and 72h. The negative and mock control groups were established.In order to test the siRNA validation, the expression of Runx2 and Galectin-3 in mRNA and protein level were examined with RT-PCR and Western Blot analyses.3. When transfected with siRNA in U251 cell, the change of cell proliferation were detected by MTT assay.4. When transfected with siRNA in U251 cell, the change of cell apoptosis were detected by FCM.Results1. Runx2 and Galectin-3 were negative in normal brain tissue, the positive expression rate of them in glioma tummor tissue was:74% and 58%.2. Runx2 and Galectin-3 were related to histological grade, tumor recurrence and 2-year survival rate (P<0.05)3. The expression of Galectin-3 was highly correlated with the expression of Runx2,there was a positive correlation between them in glioma tummor (rs=0.715,P<0.01).4. When U251 cell were treated with Runx2 Small interfering RNA, the expression of Runx2 and Galectin-3 in mRNA and protein level were brokendown,5. when use Galectin-3 Small interfering RNA treated U251 cell,only Galectin-3 were decreased in mRNA and protein level, The expression of Runx2 did not change significantly.6. Inhibit the expression of Runx2 and Galectin-3,it will result in the decrease of U251 cell proliferation activity.7. Inhibit the expression of Runx2 and Galectin-3,it will result in the increase of U251 cell apoptosis. Conclusions1. Runx2 and Galectin-3 were detected in human glioma tumor, Runx2 midiates the expression of Galectin-3.2. The specific siRNA for Runx2 and Galectin-3 can inhibit the expression of Runx2 and Galectin-3 efficiently.3. Inhibition of the expression of Runx2 and Galectin-3 can inhibit the proliferation activity in U251 cells.4. Inhibition of the expression of Runx2 and Galectin-3 can promote apoptosis in U251 cells5. Runx2 and Galectin-3 may involved in tumorigenesis and tumor progression of glioma, the detection to the expression of Runx2 and galectin-3 can be a reference valuable index for the treatment of glioma tumor.
