Objective To evaluate A medicine as a possible male contraceptive. Methods Male Kunming mice were divided into four groups of 30 each. The three experimental groups were intraperitoneal injected three different doses compared with normal saline in control group by intraperitoneal injection. Five mice of each group were sacrificed on the 7,14,21,28,35 and 42 day post injection. Testes weight,sperm count and sperm mobility in cauda epididymis, testosterone, inhibin B plasma levels and fertility were studied. Morphology of testes, A organ, B organ and C organ were analyzed with light and electron microscopy. Result Relative weight of testis (testis/body weight ratio), sperm count, sperm mobility, testosterone and inhibin B plasma levels were reduced on the 7th,14th and 21st day in all experimental groups compared with control, and fertility was notably suppressed. All the parameters recovered to normal from the 28th day. Testis showed remarkable histological changes at the 7, 14 and 21 day after vaccination, but returned to normal after 28 days. No notable morphological changes were found in the three organs. Conclusion A medicine at higher doses can inhibit male reproductive functions and fertility without substantial damage to important organs. It may have the potential to be a safe, effective and revertible male contraceptive. Objective To evaluate the effects of A medicine on spermatogenesis in rabbit, herein we give a new innovative strategy for novel contraceptive therapeutics for the male. Methods Twenty-four adult male rabbits were divided into two groups of twelve animals each; Group I -control, Groupâ…¡-injected with A medicine.7 days,14 days,21 days,28 days later, testes, epididymides, A organ and B organ were removed. Sperm count and sperm mobility were detected. Testes, A organ and B organ were analyzed for histology evaluation. They were also detected for testosterone and inhibin B plasma levels. Result On the 7th day and 14th day, sperm count and mobility showed a decline (p<0.01), A medicine inhibited the secretion of serum testosterone (p<0.01) and plasma inhibit B (p<0.01). Histological analysis of the seminiferous tubules showed that spermatogenesis was blocked at the late stages of spermatid elongation, the number of primary spermatocytes, second spermatocytes, spermatids and sperms was reduced. On the 21th day and 28th day, sperm quality and other index were unaltered. Conclusion These findings suggest that A medicine can inhibit sperm quality within two weeks, but after that, it gradually recovered.
