Abstract:Aiming at the disjunction of diagnosis and treatment of breast problems, we focus on the biological characteristics of breast cancer pathogenesis, To design and build several functional inorganic and organic nanomaterials which were most popular in current research according to the biological characteristics of breast cancer, In order to provide a new theoretical basis for nano-biomaterials study in the diagnosis and treatment of breast cancer.The main research contents and results are as follows:1. Nano grapheme was synthetized by the improved Hummer’s method, carrying photodynamic HPPH to form GO-PEG-HPPH complexes by the benzene ring structure, and compare the vivo release mechanisms and therapeutic effects of HPPH and GO-PEG-HPPH complexes in multi-modal images. Thus, it is showed that GO-PEG-HPPH complexes significantly improve the cure rate of breast cancer in the same image enhancement, and why they can reinforce the treatment efficacy of the cancer, and it is expected to change the traditional clinical photodynamic drug administration. In addition, as an effective near-infrared dye (CySCOOH) carrier, why nano-graphene enhances photoacoustic and photothermal effects is studied to laid a foundation for preclinical application of functional nano-graphite under the guidance of multi-modal images.2. nano gold morning glory was synthetized in the liquid/liquid/gas phase interface its diameter, length of the bevel edge, and thichness, range respectively in the scope of144.6±21.8nm,123.3±21.3nm±10.0±1.6nm by transmission electron microscopy (TEM). gold nanoflowers can improve their near-infrared laser absorption/scattering ratio with a special crystal structure (angular Orange Peel like flower-shaped) based on means of plasma resonance principle, and compared with gold bars, the light-heat conversion rate significantly increased (74%). Therefore, it has been by far reported to be the highest efficient solar-thermal reagent, whose photoacoustic effects are significantly stronger than the conventional photoacoustic contrast agent. The multi-phase interface may also be usesd in the chemical reactions of the fields of science and industry, such as manufacturing micro-nano crystal structure under mild conditions.3.Hman serum albumin (HSA), whose amidogen is efficiently activated by EDC/NHS system, convalently combine with CySCOOH to form a nanospheres CySCOOH@HSA whose diameter is less than lOnm of the microspheres. Compared with previous non-covalently linked HSA complexs, CySCOOH@HSA improves its biological stability, prolongs its half-life in blood, has an optical double photoacoustic imaging performance. So it passively enriches in the tumor area, and has better photothermal response performance.4. To open the cage of lactoferrin nano under acidic conditions by gradient change of PH value, parcel near infrared dye IR820. The self-assembled nano-cage will be effectively wraped up in the neutral conditions, by polymerization itself, to form a new stable optical/photoacoustic imaging materials. Aggregation of the dye molecules will produce the strong van der Waals forces, π-π Stacking adhesion, which could be obtain as light and heat treatment with fluorescent light and sound under the multi-modality imaging. The wavelength fluorescence emisses is separating to both sides. This has advantages of biphasic separation edge. A fluorescence imaging wavelength550nm has a high emission efficiency, and the808nm wavelength light, which is used into the photoacoustic image as well as heat treatment, will have high light-heat conversion efficiency.5. the nanoparticles, having an average particle size of25nm microspheres, are formed by parceling near-infrared dye in the self-assembled silane way. The nanoparticles can efficiently target there sentinel lymph node metastasis in the optical/photoacoustic imaging. Bioluminescence imaging also confirmed only in the metastatic lymph nodes which will efficiently gather nanospheres. This result shows the microstructure of metastatic lymph nodes and composition are the main reasons of these nanospheres gathered, which will provides references to the study on cancer metastasis model as a new clinical early detection.In summary, this study provides a new theoretical basis for nano-biomaterials study in the theranostics of breast cancer.
