This paper was with two polymers, β-cyclodextrin-polyethylene imine (β-CyD-PEI) and polyethylene glycol modified low generation PAMAM as a carrier material to delivery of drugs and nucleic acids, mainly divided into the following three aspects:1) A multifunctional gene carrier was synthesized successfully with redox sensitive, oligopeptide-guided, self-assembled and efficiency-enhanced, called ROSE carrier. Cytology in vitro experimental results show that ROSE carrier system can carry miR-34a into the LM3 high metastasis cancer of the liver cells, inhibiting LM3 cell proliferation and migration. In zoological experimental results showed that ROSE carrier system can efficiently carry function miR-34a to liver cancer tumor site, and can significantly inhibit the growth of liver tumor.2) The polymer PSPG was designed and synthesized with layer-invertible self-assembly properties as nano carrier materials for the first time, and was proved that the carrier can carry nucleic acids and drug molecules at the same time by the reverse of the inner and outer layer. PSPG6 had good serum stability and ability of lysosome escape, in vitro and in vivo PSPG6 with biodegradable disulfide bond structure can effectively reduce the cytotoxicity of materials, drug release and gene transfection. The treatment of DOX and Bcl-2 siRNA transported by PSPG6 can effectively inhibit the growth of the liver tumor.3) A multifunctional gene carrier was synthesized successfully with layer-invertible, oligopeptide-guided, redox-sensitive and dendrimer-based, called LORD carrier. Cytology in vitro experimental results show that LORD carrier system can carry PTX and TR3 siRNA into Panc-1 cells of the pancreatic cancer, inducing apoptosis. In zoological experimental results showed that the LORD carrier system can efficiently carry PTX and TR3 siRNA to liver cancer tumor site, inducing tumor cell apoptosis and inhibiting the growth of liver tumor.
