The blade of Porphyra yezoensis isformed by single layer of cells, and the sexual parts are differentiated from vegetativecells. There are different ability of differentiation and development in different partsof the blade and corresponding different photosynthetic activities. In order tounderstand photosynthetic process of the blade, carbon fixation abilities andphotosynthetic activities in different tissue areas have been researched. Photosyntheticvariations and responses to desiccation of vegetative and male sectors in P. yezoensisthallus have been studied. And photosynthetic mechanisms also were researched inthylakoid membrane proteomic and soluble proteomic level. The results showed that(1) there was a significantly lowest carbon fixation in the root part with the highest inthe part closing to the top. And the photosynthetic activities of the root and the topparts were relatively lower than the other parts. The contents of chlorophyll a showeda decrease tendency from the root to the top.(2) Variations in the responses of thevegetative and male sectors in P. yezoensis thallus to desiccation were compared byDual-PAM-100. The two parts showed similar tolerance to desiccation, and theformation of spermatia from vegetative cells was independent of the degree ofdesiccation. Furthermore, we proposed that desiccation tolerance of P. yezoensisperhaps was related to photosynthetic activity of PSI.(3) Thylakoid membrane, PSIand PSII were isolated from cells of P. yezoensis blades. Two-dimensionalelectrophoresis by BN-PAGE and SDS-Urea-PAGE for studying thylakoid membraneproteins in P. yezoensis thallus was established.
