Soybean is an important economic crop. In recent years, the yield and quality of soybean suffered serious damage because of the impact of diseases. To ensure yield and quality, they were important ways to understand the molecular mechanisms of disease resistance and continue to improve the disease resistance breeding with the development of molecular biology.The sumoylation in plant has demonstrated a role for pathogen defense. By bioinformatics method, we obtained all SUMO related gene in soybean. There were GmSUMO, GmSAEl, GmSAE2, GmSCE, GmSUMO Ligase(GmE3, including SIZ type, HPY2/MMS21type and PIAS-Like),GmSUMO Protea,se(including Class A, Class B1OTS type, Class B2, and Class C ESD4type), GmSUMO Domain Contein Protein, and GmSUMO Chain Binding Protein. And we analyzed chromosome location, gene duplication in evolution process and phylogenetic treeof these genes. All the SUMO system related genes express were analyzed at the level of transcription by Real-time PCRafter inoculated with Phytophthora sojael in SN10and BF25.GmSUMO2/3, GmSAElb/, GmSAE2a, an&GmSCEa/b may play a major role in root disease-resistance process.The expression of GmE3was higher than other SUMO components.especiallyGmE3/f, therefor SUMO ligase plays a vital role in the SUMO system.The expression of GmESD4α/ddecreasedsignificantly after inoculation, so theyplay a major role of the SUMO-specific protease. By western blotting of SUMO conjuction protein,theywere expressed in all tissues of the soybean,more in the root and promyelocytic.After hot shockinduction, SUMO conjuction proteins gethigh expression in soybean different tissue. The result suggested the conjugation of SUMOto substrate proteins is an important response of soybean to abioticstress. Finally,we researched the SUMO conjuction proteins express after inoculated with Phytophthora sojael.SUMO conjuction proteins have a significant increase in different tissue of SN10, but SUMO conjuction proteins reduce or slight increase in Hefeng25. Therefor the SUMO system of Soybean associated with the diffence toPhytophthora sojael.
