The apple industry was seriously affected by apple ring rot. Against the problem, in the research,the apple leaves and fruit was chosen as the test material, and the study on response of apple to the infection of B dothidea were done. And the result as follow:1. With the culture of apple leaves by spore liquid, the infected system of uninjured apple cultivated varieties to apple ring rot was improved. With the help of the system, the fast-speed identified system of apple cultivated varieties to apple ring rot was improved. The susceptible index of six apple cultivated varieties to apple ring rot was tested, and the result was same as the result of investigation outdoor. There was simple operating, short time, better repeated and non destructived test material,during the identified system improved in our research.2. Along with the time increasing of the suffering by B dothidea, the structure of the leaves chloroplast changed. The plasma membrane decomposed and disappeared, the structure of thylakoid and lamellar structure decomposed and disappeared. Because of these changes of the chloroplast structure, the function of photosynthesis and energy metabolism were reduced and disappeared.3. Compared with the other 3 percoll density gradient centrifugation methods, there was higher integrity rate, higher isolation rate, higher material utilization, that the chloroplast isolated with the help of percoll density gradient centrifugation method D. Along with the time increasing of the suffering by B dothidea, the content of chlorophyll was induced, the permeability of the plasma membrane was increased, the content of Mg2+-ATPase and Ca2+-ATPase was induced, the content of MDA, SOD, POD was improved.4. The chloroplast protein, which was extracted from apple leaves with the help of modified phenol extraction, was suitable for the analysis of 2-DE. And the improved 2-DE operating system was suit for the separation of apple leaves chloroplast protein. During the process of apple leaves chloroplast response of the ring rot pathogen stress, multiple pathways were pulsed on to achieve to pathogen stress defense, including differently associated proteins expressed in photosynthesis, energy metabolism,antioxidant response, protein metabolism, amino acid metabolism.5. The results showed that the modified phenol extraction method was suit for the extraction of apple fruit skin protein, and the improved 2-DE operating system was suit for the separation of apple fruit skin protein. During the process of apple fruit skin response of the ring rot pathogen stress,multiple pathways were pulsed on to achieve to pathogen stress defense, including differently associated proteins expressed in energy metabolism, saccharometabolism, protein metabolism, amino acid metabolism, defense and stress response.
