| [Object] Recent experimental work has demonstrated that five muscarinic acetylcholine receptor (mAChR) subtypes (ml-m5) are expressed in the vestibular periphery. However, the expression of the mAChRs protein in the vestibular periphery of guinea pig were unclear. In this study, we investigate whether there is the expression of M1-M5 muscarinic receptor protein in vestibular end-organ (including the posterior ampulla, horizontal and superior ampulla, utricle, saccule) of guinea pig.[Methods] Using western blot to detect the expression of M1-M5 muscarinic receptor protein in vestibular end-organ (including the posterior ampulla, horizontal and superior ampulla, utricle, saccule) of guinea pig.[Results] In vestibular end-organ(including the posterior ampulla, horizontal and superior ampulla, utricle, saccule), protein of the expected size was obtained by western blot.[Conclusion] These results demonstrate the expression of M1-M5 mAChR protein in the the posterior ampulla, horizontal and superior ampulla, utricle, saccule of the guinea pig, and lend further support for the role of M1-M5 may be one of the mediators of efferent cholinergic signaling in vestibular hair cells as well. [Object] Muscarinic acetylcholine receptors (mAChRs) are widely expressed in the central and peripheral nervous systems and play an important role in modulating the cell activity and function. However, the expression of M1-M5 muscarinic receptor mRNA in vestibular end-organ of guinea pig was unclear. In this study, we investigate the expression of the five muscarinic acetylcholine receptor (mAChR) subtypes (m1-m5) in vestibular end-organ of guinea pig.[Methods] Using reverse transcription polymerase chain reaction (RT-PCR) technique to detect the expression of M1-M5 muscarinic receptor mRNA in vestibular end-organ of guinea pig.[Results] In vestibular end-organ, M1-M5 mRNA was identified by RT-PCR. The products were confirmed to be M1-M5 by sequence analysis.[Conclusion] These results demonstrate the expression of M1-M5 mAChR in vestibular end-organ of the guinea pig, and lend further support for the role of M1-M5 may be one of the mediators of efferent cholinergic signaling in vestibular hair cells as well.
