| Background and ObjectivesBladder cancer, with the highest morbidity and mortality among the urinary system tumors, is one of the most common malignant neoplasms of the urinary system in China, which threatens people’s life and health severely. The clinical treatment strategies for bladder cancer is mainly surgical excision, combined with infusion chemotherapy and immunotherapy treatments, which has unsatisfactory results. Find the incidence of bladder cancer gene to understand the molecular genetics of bladder cancer, and provide a theoretical basis for the diagnosis and treatment of bladder cancer is the research focus of the current.Numb is an evolutionary conserved protein that plays critical roles in cell fate determination. Numb proteins display a complex pattern of functions such as the control of asymmetric cell division and cell fate choice, endocytosis, cell adhesion, cell migration, ubiquitination of specific substrates and a number of signaling pathways (i.e. Notch, p53). The alterations of Numb expressing have been implicated in tumorigenesis, but its influence on the biological behavior of a bladder cancer cell is still unknown. We hope to clarify the possible molecular mechanisms of Numb in Hes1, p53pathway on invasive bladder cancel SO as to provide new strategies on early diagnosis and treatment of bladder Cancel.Methods (1) The expression of Numb was detected by qRT-PCR and Western-blot in20cases of bladder cancer, adjacent non-rumor tissues. Three bladder cancer cell lines(T24, J82and5637and bladder epithelial immortalized cell(Sv-HUC-1))-The relationship of Numb with clinical stage and pathological grade was also studied.(2) After transfection with the plasmid containing the ORF clone of Numb, The effects of Numb on cells proliferation, apoptosis, were detected by MTS assay and flow cytometry analyses.(3) The alteration of Numb, hairy and enhancer of split1(Hesl), p53in5637ã€J82ã€T24were analyzed by western blotting and qRT-PCR, before and after transfection with the plasmid containing the ORF clone of Numb.Results:The expression of Numb was down-regulated in20samples compared with paired adjacent non-tumor tissue. According to the clinical stage, the relative expression level of Numb was not significant difference between T2and T3-T4(P>0.05). According to the pathological grade, the relative expression level of Numb in well-differentiated group was significantly higher than that in low-moderately differentiated group. The Numb expression in normal bladder cell line SV-HUC-1was significantly higher than that in the three bladder cancer cell lines (T24, J82and5637)(P<0.05), and it gradually decreased(5637>J82>T24) with increasing degree of malignancy of bladder cancer cells. The overexpression Numb resulted in the attenuation of proliferation, with Hesl down-regulation in J82and T24. The5637cell line had an opposite effect. The expression of p53in the three cell line was not significant difference after transfection with the plasmid containing the ORF clone of Numb. Conclusions:Our findings indicated that Numb expression was down-regulated in low-moderately differentiated invasive bladder cancer, and that it could regulate Hesl expression through the Notch pathway. These might provide a basis for the designing new therapeutic strategies for invasive bladder cancer. |
