Objective:To observe effect of Fuzheng Yiliu decoction on tumor cells apoptosis and expression of Beclin1, Bnip3and LC3of human hepatoma cells-Induced nude mice model, and investigate the molecular mechanisms of Fuzheng Yiliu decoction inhibiting tumor.Methods:Nude mice model of tumor was established by transplantation of human hepatocellular carcinoma (Hu-7and HepG-2). Nude mice models of Hu-7and HepG-2were distributed fifty respectively. Two kinds of nude mice model were allocated randomly to five groups. There were normal saline group (blank control group, BC), high dose group of Fuzheng Yiliu (FZYLH), middle dose group of Fuzheng Yiliu (FZYLM), low dose group of Fuzheng Yiliu (FZYLL) and5-Fu group,10nude mice per group. Intervention was administered in each group when tumor was size of100mm3. Fuzheng Yiliu groups were administered Fuzheng Yiliu decoction by lavage according to fixed concentration, once daily.5-Fu group was administered5-Fluorouracil (5-Fu) via intraperitoneal injection, twice a week, while NS was administered saline via intraperitoneal injection, once daily. After intervention of18days, nude mice were sacrificed and tumor was severed, weighed up. We employed tunnel dyeing to evaluate tumor cells apoptosis, transmission electron microscope (TEM) to assess formation of cytophagosome, and western blot to detect the expression of Beclin1, Bnip3and LC3. These results were approached by statistics with software of PAS W statistics18.0.Results:Ratio to inhibitory tumor:Fuzheng Yiliu decoction and5-Fu could inhibit growth of tumours. FZYLH and5-Fu has an advantage over the control group (P<0.05), while FZYLL has little effect on growth of inhibitory tumours (P>0.05). Ratio to inhibitory tumor was49.9%,21.6%,8.2%, and74.2%respectively in Fuzheng Yiliu decoction high, midle, low dose groups and5-Fu group.Tumor cells apoptosis:A lot of tumor cells apoptosis were perceived in FZYLH and5-Fu compared with BC. HDFY has an advantage of tumor cells apoptosis over BC (P<0.05), while there was no significant difference among FZYLM, FZYLL and BC (P>0.05).Associated protein of autophagosome:TEM result showed that there were a lot of autophagosome in Fuzheng Yiliu group and5-Fu group, and there was a positive correlations between the quantity of autophagosome and drug concentration. Westen-blot analysis showed that expression of Beclinl, Bnip3and LC3was up-regulated in Fuzheng Yiliu group and5-Fu group, and there was still a positive correlations between their expression and drug concentration.Conclusion:These results demonstrated that (1) Fuzheng Yiliu decoction could inhibit the growth of tumor cells in nude mice, up-regulate expression of Beclin1, Bnip3and LC3proteins, and induce tumor cells apoptosis and autophagy,(2) activation of autophagy would be one of the acting mechanism of anti-tumor of Fuzhengyiliu decoction. |
