Effects Of The Drug Of Chongcaoyishen Particles On The Nephrin And Podocin Podocyte Cells Of Membranous Nephropathy Rats

Objective:Through observe the protective effect of chongcaoyishen granule on sertoli cell and renal function of rats with membranous nephropathy, meanwhile discuss the mechanism of action about treating membranous nephropathy and delaying the renal fibrosis.Methods:Select 72 clean healthy male SD rats, randomly selected 12 rats for blank group,the rest of the mouse tail vein once every other day was injected cation serum albumin (C-BSA) to copy membranous nephropathy (MN)rats model.After the success of the building,the remaining rats were randomly divided into 5 groups respectively were model control group,western medicine control group (GC+CsA),the low—dose group of Chongcaoyishen medicine, medium—dose group of Chongcaoyishen medicine, high-dose group of Chongcaoyishen medicine, Dosage regimen:blank group and model control group were given the same amount of distilled water to fill the stomach, western medicine control group was given hormone and ring spore sodium chloride solution to fill the stomach, the low—dose group,medium—dose,high-dose of Chongcaoyishen medicine were given chongcaoyishen granule water decoction to fill the stomach,once a day,for four consecutive weeks,before building,before drug administration, two weeks after the treatment,four weeks after the treatment respectively, observed in 24—hour urine protein of rats, the last time after dosing, then will kill all rats, detect rats in different groups changes of serum total protein(TP),albumin(ALb),total triglyceride(TG),urea nitrogen (BUN),serum creatinine (Scr),return of renal tissue in rats,pathological changes of renal tissues of rats were observed by immunofluorescence, light microscopy and electron microscopy. The expression of Nephrin mRNA, podocin mRNA, PAI-1mRNA, FN mRNA, LN mRNA, in the renal tissues were tested by immunohistochemistry and Real-time PCR.Results:1. Chongcaoyishen granule improve the general state of MN rats, increase the level of TP, Alb, reduce the 24 hour urine protein of MN rats, reduce T-CHOL,TG, BUN,Scr,compared with the model control group with significant difference(p<0.05,p<0.01),the middle group of Chongcaoyishen granule was best,compared with the hormone and ring spore group,there was no significant difference(p> 0.05).2.Chongcaoyishen granule reduce the renal pathological damage of MN rats, increase the expression of Nephrine, prodocin in the renal tissues, compared with the model control group with significant difference(p<0.05), and the middle dose of chongcao yishen capsules have the most obvious rising, compared with the hormone and ring group,there was no significant difference(p> 0.05).3. Chongcaoyishen granule can restrain the expression of PAI-1, FN, LN in renal tissues,compared with the model control group with significant difference(p<0.05,p<0.01), and the middle dose of chongcao yishen capsules to reduce the most obvious,compared with the hormone and ring group,there was no significant difference(p> 0.05).4. Chongcaoyishen granule can increase the expression of Nephrine mRNA, prodocin mRNA in renal tissues of MN rats,compared with the model control group with significant differenc(p<0.01), and the middle dose of chongcao yishen capsules to reduce the most obvious,compared with the hormone and ring group,there was no significant difference(p> 0.05).5. Chongcaoyishen granule can reduce the expression of PAI-1 mRNA, FN mRNA, LN mRNA in renal tissues of MN rats,compared with the model control group with significant difference(p<0.05,p<0.01), and and the middle dose of chongcao yishen capsules to reduce the most obvious,compared with the hormone and ring group,there was no significant difference(p> 0.05).Conclusions:1.chongcaoyishen granule can improved MN rats of TP,Alb decline the 24hour urine protein,decline T-CHOL,TG, improve BUN, Scr.2.Chongcaoyishen granule may be through increasing the expression of Nephrine mRNA, prodocin mRNA in renal tissues of MN rats,to improve sertoli cell in the integrity of the membrane structure,to protect sertoli cell.3. Chongcaoyishen granule may be through revising the expression of PA1-1 mRNA, FN mRNA, LN mRNA in renal tissues, accelerate the ECM degradation,reduce the accumulation of ECM, relieve renal fibrosis, delay the progress of membranous nephropathy.4. Chongcaoyishen granule can reduce membranous nephropathy rats renal pathological damage,inhibition of membranous nephropathy rat glomerular basement membrane immune complex deposition and the thickening of basement membrane.delay the progress of membranous nephropathy.5. Chongcaoyishen granule compared with hormone and ring, the low—ose group, medium—dose group of, high-dose group of Chongcaoyishen medicine and hormone and ring group can protect the kidneys to some extent, relieve renal pathological injure of MN rats, inhibit the deposition of immune complex in membranous nephropathy glomerular basement membrane and thickening of basement membrane of MN rats.
