Objective: The aim of this study is to investigate the effect of adipose-derivedstem cells (ADSCs) transfected with chondromodulin (ChM) on articular cartilagedefect in rabbit. Methods: ADSCs were transfected with green fluorescent protein (GFP)labeled lentiviral vector which contained ChM-1or control vector. GFP positive cellswere separated by flow cytometry. Alizarin red staining and oil red O staining wereperformed for multi-directional differentiation determination. ChM expression in thecells was assayed by western blot and real time PCR. Eighteen rabbits were randomlydivided into3groups: bone defect group, control group, and ChM-1group. ChM-1transfected ADSCs were inoculated onto the scaffold and cultured for24h. Surgicalbone defect on the knee were made and the scaffold contained ADSCs with or withoutChM-1transfection were implanted. After12weeks, the rabbits were sacrificed and theknee was fixed for the sequential detection. Morphology of the cartilage was observedand the repair effect was evaluated by toluidine blue staining of type II collagen andWakitani scoring. Results: Increased expression of ChM-1was observed in ChM-1gene transfected ADSCs. Compared with the control group, the defect site of the ChM-1group has been connected to the adjacent normal cartilage12weeks after surgery.Toluidine blue staining and Wakitani scoring showed significant advantages of theChM-1group on cartilage repair compared with other groups.Conclusion: ChM-1transfected adipose-derived stem cells could play a conclusive effect on articularcartilage defect.
