| Objective: 1) to explore the distribution of the genetic polymorphisms of four locies such as rs1800805, rs1800807, rs1800808 and rs6136 in thrombosis formation-related genes, P-selectins(Ps), in among Han Chinese, Uygur and Kazakh populations of Xinjiang China and their potential relationship with atrial fibrillation(AF) in these three ethnic groups; 2) to explore the potential relationship between plasma concentrations of soluble P-selectin(sPs), related to thobosis formation and Ps, and AF coupled with thrombosis; 3) to further analyze the relationships of genetic polymorphisms of PS and plsma concentrations of sPs, based on the results of genetic polymorphisms and thrombosis in atrial fibrillation. Methods: 1) 690 Han Chinese subjects,(121 patients with atrial fibrillation and with thrombosis, 219 patients with atrial fibrillation and 350 normal subjects without atrial fibrillation and thrombosis), 562 Uygur subjects(103 patients with atrial fibrillation and with thrombosis, 199 patients with atrial fibrillation and 260 normal subjects without atrial fibrillation and thrombosis), 505 Kazakh subjects(comprised of 42 patients with atrial fibrillation and with thrombosis, 156 patients with atrial fibrillation and307 normal subjects without atrial fibrillation and thrombosis) were enrolled into this study, subdivided into three groups as atrial fibrillation coupled with thrombosis(case group), atrial fibrillation not coupled with thrombosis(control group) and non atrial fibrillation and non thrombolism(normal group) and matched in terms of age and gender.PCR-RFLP methods were performed to analyze genetic polymorphisms of rs1800805,rs1800807, rs 1800808 and rs6136;2) ELISA method was used to measure plasma concentrations of soluble P-selectins; 3) relationships of polymorphisms of PS gene and plasma concnetrations of soluble P-selectins, based on the results of above measurements.Results:1) polymorphisms of rs1800807 and rs1800808 were significantly correlated withthrombosis in atrial fibrillation among Han Chinese, Uygur and Kazakh subjects enrolled into this study. GG genotype of rs1800807 and TT genotype of rs1800808 were observed to increase the development of thrombosis in atrial fibrillation among Han Chinese, Uygur and Kazakh subjects in this study, even after adjusting for related confounders. GG genotype of rs1800807 and TT genotype of 1800808 showed significant positive correlations with the plasma concentrations of soluble P-selectins in Han Chinese, Uygur and Kazakh subjects of Xinjiang China in this study. The locies as rs1800805 and rs6136 showed no significant genetic polymorphisms in Han Chinese, Uygur and Kazakh subjects of Xinjiang China. 2) No significant differences were observed in plasma levels of soluble P-selectin among three ethnic groups, whereas significantly correlated to AF coupled with thrombosis in these ethnic groups. 3) Plasma levels of soluble P-selectin were significantly higher in carriers of GG genotype of rs1800807 and of CT and TT genotypes of rs180080 loci.Conclusion: GG genotypes of rs1800807 and TT and CT genotypes of rs1800808 locies might increase the risk of developing thrombolism in AF and might elevate the levels of soluble P-selectin, further accelerating development of thrombosis, whereas genotypes of PS gene and levels of soluble P-selectin might not have significant differences among these three ethnic groups. |
