Objective:The Experimental research is designed to observe the influence of the supplemented San-Cai decotion on the liver fatty degeneration and keap1-Nrf2 transduction of the rats with diabetes mellitus, and study the mechanisms. It will provide the scientific basis for that the supplemented San-Cai decotion’s becoming a new Chinese patent drug.Methods:90 healthy male SD rats were randomly divided into normal control group(10 rats, fed with normal food) and experimental group(80 rats, fed with high-sugar and high-fat food). Five weeks later, streptozotocin(STZ 30mg/kg) was injected intraperitoneally to esablish experimental diabetic rat models. Blood glucose levels were measured (>16.7mmol/L) on the third day after STZ injection to confirm the development of diabetes. Then the diabetic rats(70 rats) were subdivided randomly into five guoups:DM model group(12 rats), the groups of supplemented San-Cai decotion in low dose (15 rats), middle dose(15 rats) and high dose(13 rats), and the contrast group of Shuilinjia(15 rats, drenched Shuilinjia suspension 28mg·kg-1·d-1). In the five groups, except the model group, others were treated by corresponding medicine. After 4 weeks, by the end of the experiment, all rats were put to death, and the following items were detected:the weight of rats; the serum contents of TC, TG, HDL, LDL, free fatty acids (FFA); the serum contents of fasting blood glucose (FBG)and fasting insulin (FINS); The serum contents of malondialdehyde (MDA) and hydrogen peroxide(H2O2);the activity of kelch-like ECH-associated protein 1 (Keap1), nuclear factor-E2-related factor 2 (Nrf2), superoxide dismutase (SOD), catalase(CAT) and Glutathione peroxidase (GPX) were detected in the liver tissue homogenate. The liver histopathology was observed by hematoxylin and eosin (HE) staining.Results:The supplemented San-Cai decotion could reduce the liver fatty degeneration. After to analyze the experimental data, we found that, all above indexes in model group are abnormal compared with those in normal group (P<0.05, P<0.01), but the indexes are all improved in treated groups than those in model group. The indexes of TC, TG, LDL, FFA, FBQ INS, and MDA are all improved better in all treated groups than those in model group (P<0.05). Compared with model group, the level of H2O2 and Keap1 in the hepatic tissue obviously reduced in High and middle dose groups (P<0.05); Compared with model group, the level of HDL, Nrf2 and its gene expression and SOD’s gene expression obviously increased in High and middle dose groups (P<0.05); Compared with model group, the level of CAT and GPX in the hepatic tissue obviously increased (P<0.05); but Keap1’s gene expression obviously reduced in High dose group (P<0.05);Conclusion:1. Male SD rat model of diabetic fatty liver was established with high-fat diet for 9 weeks and intraperitoneal injection of Streptozotocin (30mg/kg). This model accompanied with hyperglycemia, hyperinsulinmia and insulin resistance is similar to type 2 diabetes of human. It is a suitable model to study the NAFLD of type 2 diabetes mellitus.2. The supplemented San-Cai decotion could improve insulin resistance through reducing the level of FBG,INS, and correcting disordered blood lipids metabolism.The supplemented San-Cai decotion could increase the ability of antioxidation, through improving the activities of Keapl-Nrf2 and antioxidant enzymes (including SOD,CAT and GPX). These effects may be the mechanism of the supplemented San-Cai decotion’s protection of the liver.
