Objective:To evaluate the effects, safety and related mechanisms of estrodiol and isopropanolic cimicifuga racemosa extract on symptoms releasing and vessel protection in ovariectomized mice.Methods:A total of 20 female C57BL/6 mice and 20 female APOE-/-mice were randomly divided into the following groups:surgery with vehicle (SHAM+NS), ovariectomy with vehicle (OVX+NS), ovariectomy with E2 treatment (OVX+E2), and ovariectomy with Remifemin (OVX+ICR). The weight changes, the serum lipid spectrum, the aortic atherosclerosis areas, the reproductive system and the nervous system were observed after 12 weeks of treatment.Results:Compared with the SHAM+NS and OVX+E2 group, the body weight, abdominal fat, serum cholesterol and aortic atherosclerosis plaque areas of the OVX+NS and OVX+ICR were increased, while the weight of uterus decreased. In the tail suspension test, the immobility time of SHAM+NS, OVX+E2, OVX+ICR group is shorter than the OVX+NS (P< 0.05).Conclusions:The estradiol could improve the serum lipid spectrum, decreases the body weight, abdominal fat and aortic atherosclerotic plaque areas of the ovariectomized mice, while the remifemin didn’t. The estradiol could promote the proliferation of the uterus; while the remifemin might not. Both of the estradiol and remifemin could shorten the immobility time of the ovaryectomized mice in the tail suspension test, thus improved the mental status.
