Objectives:To observe the enviromental impact of renal aging through establishing Young-Old rats kidney transplanted model; Using non-labeled quantitative proteomics method to screen the protein associated with aging, then verify it and explore the possible mechanism of the impact of aging.Methods:â‘ Through in situ kidney resection and donor kidney transplantation method, the young rats kidney transplanted to old rats, or the rats kidney transplantation to the young rats. After in vivo growth 6 months after, remove the kidney have been transplanted. â‘¡Observe the kidney pathology (HE, PAS staining), tissue senescence (β-gal staining), and inflammator cytokines (TNF-αå'Œ IL-1β) after transplantation; â‘¢ Using label free proteomic analysis to screen protiens involved in renal aging, ahd validate it in Wi38 cells.Result:â‘  We established Young-Old rats kidney transplanted model successfully; â‘¡ Compared with kidney from young rat, we found no changes in HE staining and PAS staning, increasing β-gal staining, and increasing expression of TNF-αå'Œ IL-1 β in young kidney transplanted to old rat; Compared with kidney from old rat, we found no changes in HE staining and PAS staning, decresing β-gal staining, and decreasing expression of TNF-αå'Œ IL-1 β in old kidney transplanted to yong rat; â‘¢ A strict cutoff value of a 2.0-fold change resulted in a final set of 279 differentially expressed proteins, and 16 protains associated with ATP energy metabolism/apoptosis/cell death, and protein-protein interaction analysis showed that Sod1 was a multiple interacting protein. Compared with 20 generation Wi38 cells, the expression of Sod1 was decresing and the expression of p16 was increasing in 40 generation Wi38 cells. Ac-NF-κB expression decreased when Sodl over-expression, while Ac-NF-kB increased when Sod1 knocked out.Conclusion:1. We established Young-Old rats kidney transplanted model successfully;2. The environment may affect the renal aging, in the Young-Old rats kidney transplanted model, environment could make old kidney younger/young kidney oldner.3. Protenmic analysis showed that Sod1 invovled (inhibited) renal aging, and inhibit NF-κB pathway to inhibit aging.
