| Objective:In this paper, we trying to investigate the absorbed components of Danggui-Shaoyao-San (DSS) in rat plasma and their pharmacokinetics.Methods:In this paper, HPLC-MS/MS and UPLC-MS/MS methods were developed for the qualitative and quantitative analysis of the major constituents in DSS. The MS fragmentation behavior of the reference compounds was proposed for aiding the structural identification of components in DSS extract. The linearity, precision, accuracy, stability were validated for the quantification method, which proved sensitive, accurate and reproducible and further-more apply to comparative pharmacokinetics of them after oral administration of pure BDPH, ALI and DSS extract. The pharmacokinetic parameters were calculated by DAS 2.0 software, and the pharmacokinetic parameters were compared in each group.Results:40 compounds were identified or tentatively characterized from DSS by comparing their retention times, UV and MS spectra with those of authentic compounds or literature data, and 22 of them were determined in rat plasma by HPLC-MS/MS.34 compounds were identified from DSS and 25 of them were determined in rat plasma by UPLC-MS/MS. Senkyunolide A, Z-Butylidenephthalide, Z-ligutilide, Alisol B, Albitlorin, patonitlorin, Atractylenolide â…¢, Atractylenolide â…¡, Pachymic acid, Catechin might be the main absorbed compounds. BDPH and ALI in rat plasma were determined by HPLC-MS/MS, and main pharmacokinetic parameters were calculated. Pharmacokinetic comparative study showed that the absorption of BDPH and ALI in DSS extracts was all significantly increased.Conelusion:Senkyunolide A,Z-Butylidenephthalide,Z-ligutilide, Alisol B,Albitlorin, patonitlorin, Atractylenolide III, Atractylenolide II,Pachymic acid,Catechin might be the main absorbed compounds. A sensitive and specific high-performance liquid chromatography-electrospray ionization-tandem mass spectrometry method was developed to analyze BDPH and ALI in rat plasma. The pharmacokinetics of BDPH and ALI after oral administration of DSS extracts was compared to evaluate the effect of purity of DSS. Pharmacokinetic comparative study showed that the absorption of BDPH and ALI in DSS extracts was all significantly increased. |
