Objective:Construct a experimental animal models similar to human skin aging process, and verify the role and mechanism of leptin in delaying skin aging process from the oxidative stress theory of skin aging. So as to finding experimental and theoretical basis for leptin in skin anti-aging and even regenerating damaged skin.Methods:Simulating factors inducing aging of human skin, build a suitable animal models for skin aging by combining D-gal and UV irradiation. Histomorphology observation and tissue homogenate oxidative stress indexs were recorded and comparative analyzed. Three leptin dose group (5ug/g、10ug/g、 20ug/g) were given by intraperitoneal injection. Histomorphology observation and oxidative stress indexs were detected after 6 weeks. UV irradiation can induce human skin fibroblasts (HSFs) aging. Various concentration (5、10、20μg/ml) of leptin solutions were administratived to HSFs in advance to UV irradiation. HSFs vogor,SA-β-gal, oxidative stress indexs and protein expression of p67phox,PKC ε,p66Shc were measured and analyzed.Results:UV irradiation of 611.28mJ/cm2 once other other day combining with D-gal of 1mg/g/D per day subcutaneous injected can lead to changes of skin histomorphology in 6 weeks. So does the oxidative stress indexs. Administration systemically of leptin can relieve histomorphology changes of aging, improve the steady of oxidation and anti-oxidation. Our study also find different concentration of leptin takes different action in regulation of oxidative stress. Leptin can alleviate the reduction of UV on antioxidant enzyme activity of HSFs and the damaging of membrane lipids and collagen, and reduce effectively the increasing expression of fibroblast p67phox, PKCε and p66Shc induced by UV radiation.Conclusion:Adipokine Leptin is a pleiotropic hormone. The study found that both in animal and cellular level, leptin can play a role in anti-oxidation, alleviating histological and cytological changes of skin aging. The mechanism may be related to the scavenging of reactive oxygen species, and maintaining the balance of oxidative stress.
