Objective TO study the level of oxidative stress on obese children and the relationship between oxidative stress and fat metabolism, TO further study the regulation of oxidative stress on the metabolize of adipose.Methods 62 cases children with simple obesity were chosen in our study as obesity group. 6-12 years old(average 9.12±3.23 years), male 36 and female 26 cases. Patients with known diseases related with heredity,endocrine metabolism and central nervous system were excluded. At the same time, 46 cases healthy children as control group,also 6-12 years old(average8.78±2.94 years),male 26 and female 20 cases.After being fasted for 10 hours,they were measured the serum triglyceride(TG), total cholesterol(TC), high density lipoprotein cholesterol(HDL-C), low density lipoprotein cholesterol(LDL-C), superoxide dismutase(SOD),8-isoprostaglandin F2α(8-iso PGF2α), Fasting insulin(FINS), Fasting blood glucose(FBG), Tumour necrosis factor-alpha(TNF-a),Interleukin-6(IL-6), C-reactive protein(CRP). Moreover we measured their blood pressure, height, weight and calculated the body mass index(BMI).SPSS 13.0 software was used to analyze data. One-way analysis(ANOVA)and Line correlation analysis was assessed, Statistical significance was accepted at p<0.05.Results1.The level of 8-iso PGF2α is higher and SOD is lower in serum of obese group than the control group(p<0.01);the level of TNF-a、IL-6、CRP is higher(p<0.01 or 0.05); Insulin and HOMR-A is higher(p<0.01 or 0.05);FBG is slightly higher(p<0.05);25-(OH)D is lower and leptin is significantly higher(p<0.01).2.42 cases of vitamin D deficiency is in obese group, 10 cases in control group,the deficiency of vitamin D in obese group is significantly higher than control group,x2=22.384(p<0.01).3.36 cases of HOMA-IR is in obese group. 8-iso-PGF2a、TNF-a、IL-6、CRP and BMI、leptin is higher in HOMA-IR than no HOMA-IR(p<0.01),25-(OH)D、SOD is lower(p<0.01).4.8-iso-PGF2α is positively correlated with leptin 、 BMI and TNF-a.the correlation coefficient respectively are 0.754;0.389;0.952,the difference is significant(p<0.01), and is negative correlated with 25-(OH)D,the correlation coefficient is-0.868,the difference is significant(p<0.01). Multiple linear regression model was built about fat metabolic makers to 8-iso-PGF2α.8-iso-PGF2α =10.511-0.136[25-(OH)D]+1.15TNF-a.Conclusion The level of OS was improved in obese children.and correlated with the metabolic of adipocyte. the level of vitamin D is lower,Leptin resistance is exist and the inflammatory cytokines increased in obesity.Therefore,improved the ability of anti-oxidation and supplement of Vitamin D probably reduce the complication and incidence of obesity.
