Objective:To observe the effects of damage and repair of rats gastric mucosal with electroacupunture at Sibai (ST 2), Liangmen (ST 21), and Zusanli (ST36) in the Zuyangming Meridian, and explore its related protein molecules with relationship of the acupuncture.Methods:the gastric mucosal damage model in rats was obtained by ethanol gavage. Electro-acupuncture at Sibai (ST2), Liangmen (ST21) and Zu-sanli (ST36) of the Foot-Yangming meridians on gastric mucosal injury in rats 10 days treatment. Use pronase digestion method to isolate gastric mucosal cells, Ultrasonic fragmentation method to extract protein, to obtain gastric mucosal cell protein patterns of blank group(BGR), model group(MGR), acupuncture treatment group (ATGR) and acupuncture Control group(ACGR) rats using two-dimensional gel electrophoresis technique. By comparing the differences of protein expression levels, Choose meaningful points to do mass spectrometry. The protein finger printing of rat gastric mucosal cell for NGR, MGR, ATGR and ACGR were obtained by surface enhanced laser desoiption/ionization time of fligt-mass-spectrometry (SELDI-TOF-MS) and With WCX2 protein chip. Thereby identify the specific correlated protein signaling molecules of electric acupuncture effect,to further illuminate complicated network regulatory mechanism to provide the theoretical basis. Results:1. By comparison, the gastric mucosa lesion index in MGR rats is the highest, but the one in BGR rats is the lowest, Comparison between the two have significant differences (p<0.05), Model proved to be successful; Compared with the MGR rats, the gastric mucosa lesion index in ATGR and ACGR rats are significantly lower,which has significant difference (P<0.05); The gastric mucosa lesion index in ATGR rats iS decreased By comparison of ACGR rats, which has significant (P<0.05)2. The total of the significant difference of 7 protein spots are found out by comparing BGR rats, MGR rats, ATGR rats and ACGR rats (P<0.05).There are a total of six proteins identified through mass spectrometry.3. Compared with BGR rats,the MGR rats have a total of four proteins that are significantly different (P<0.05),2 of which obviously are upgraded, other obviously are degraded; Compared with the MGR rats, there are 4 significant differential protein peaks for ATGR rats (P<0.05),one only is upgraded,other are declined; compared with the MGR rats, there are 5 significant differential protein peaks for ACGR rats (P<0.05),2 is increased,3 is decreased; There are a total of 12 significant differential protein peaks in comparison with ACGR rats,which are manifested as increased.4. There are two protein peaks that are obviously different,which are 10777.71、3727.126, which may Specific effects of substances of gastric mucosa injury and repair in rats by electric-acupuncture treatment.5. From the experimental results,it is mainly distributed in the middle and high molecular weight protein through the two-dimensional gel electrophoresis- mass spectrometry to identify.And that is mainly concentrated in low molecular weight protein abundance through the SELDI-TOF-MS protein chip technology to identify.So the two results did not find the same or similar molecular weight differences in protein.Conclusions:1. Electro-acupuncture at the acupoints of the Stomach Meridian could have repairing effect on gastric mucosal lesion in stress-induced rats, which Prompted there is a relative particularity between Foot-Yangming meridian and stomach.2. The identified 6 proteins could, respectively, through a variety of the role of access, such as apoptosis, exert damage and anti-damage repair role. Electro-acupuncture can increase or decrease the quantity of these proteins, it is possible to play a treatment role by adjusting these proteins.3. The significant differential protein peaks can be found for cell protein finger printing founded by comparison with the BGR rats、the MGR rats、ATGR rats and ACGR rats,which is there specific significance for us to look for differential biologic signal protein molecule,which provides theories basis for the mechanisms of gastric mucosa injury and repair in rats by electro-acupuncture treatment.
