BackgroundCervical cancer is a kind of happened at cervical intraepithelial malignancies, but also the world, second only to breast cancer, a serious threat to women’s health and lives of malignant tumors. Its incidence in our country axounted for the top female malignancy, according to World Health Organization estimates that every year 131,000 new cases, accounting for the world’s new cases of cervical cancer of 28.8%.Women of any age can develop cervical cancer. However, it frequently strikes-and sometimes kills-women when they are relatively young,in their mid-to-late 40s. In recent years, young cancer patients have a marked increase in the trend to retain reproductive function and improve quality of life is imperative Cervical cancer occurs in low socio-economic status of women, and sexual health possible, HPV infection, early marriage, smoking and so on. Developing countries the incidence of cervical cancer is 6 times that of developed countries, but also because of early cervical cancer screening is not yet universal,80% of the pati ents at the time of di agnosi s for invasi ve card noma has developed.Early cervical cancer is currently mainly used for surgery or radiation therapy for advanced as well as the recurrence and metastasis of many cancer patients using radiation therapy combined with systemic chemotherapy treatment. Clinical findings, between different patients, the differences in the sensitivity of radiotherapy and chemotherapy, resultin in therapeutic effects are also very different. Improve the dose of radiotherapy and chemotherapy at a certain extent, although that can enhance the effectiveness, but different tolerance to different patients, and a marked increase in toxicity,in pati ents with severely reduced quality of life.Hedyotis diffusa is a herb plant of one year of Rubia cordifolia species which has many function such as clearing up heat and detoxicating, activating blood circulation to dissipate blood stases, urinating and so on. Modern study indicated that the main chemical composition of hedyotis diffusa have ursolic acid, immunopoly saccharide and hedyotis diffusain. They can strengthen immune activity and resist tumor activity. Clinic observation fund out hedyotisdiffusa has prominent efficacy to cervical cancer. Ki-67 is high expression in cervical cancer and other tumor. The expression of such high status and the incidence of tumor development, recurrence, as well as generated by radiotherapy and chemotherapy resistance are closely related to. Objective: To study the principle of using animal serum to study the Hedyotis diffusa on cervical cancer cell proliferation and apoptosis and Ki-67 gene expression to explore the inhibition of Hedyotis diffusa on cervical cancer and its possible mechanism of molecular biology.Method1.Use of human cervical cancer Hela cells,21,36,72 hlater, MTT cell proliferation assay, TNUEL cell apoptosis detected using RT-PCR detection of Ki-67mRNA expression of immunohistochemical detection of Ki-67 protein expression in.2. To establish nude mouse transplantation model of human cervical cancer cells and gastric perfusion. Hela cells were obtained from female,4 to 6-week-old BALB/C/nu nude mice subcutaneous tissue in the back. When the tumor growth to 10mm in diameter were randomly divided into two groups, control group:untreated tumor-bearing mice, to continue feeding into the end of the experiment; Treatment group:Hedyotis diffusa decoction gavage, then killed all the animals get tumors were weighed to calculate the inhibition rate of tumor lines immunohistochemical staining Ki-67 expression, TUNEL assay of apoptosis.Results1.Hedyotis diffusa inhibitory effect on cervical cancer cells and induces apoptosis in Hela cells, Ki-67 gene expression decreased in a dose-and time-dependent.2. For Hela cells significantly inhibited tumor growth and induce apoptosis in Hela cells, Ki-67 gene expression decreased. ConclusionsOldenlandia diffusa on cervical cancer Hela cells significantly inhibited the induction of apoptosis, and inhibit the expression of Ki-67 gene, which may inhibit tumor growth of its molecular biological mechanism.
