Aim:The present study aims to investigate whether the traditional Chinese medicine (TCM)——Baicalin, Puerarin and Ginsenoside Rgl, could influence the expression level of miR-290cluster thereafter to regulate the proliferation of mouse D3embryonic stem cells (ESCs).Methods:In the study, we investigated the proliferation of ESCs by calculating the cell population double time (Td) through the cell counting and detecting the cell cycle using PI staining. The expression of proto-oncogenes (c-myc, c-jun and c-fos) and the expression of members of miR-290cluster were detected by real time quantitative PCR. Moreover, the miR-294was overexpressed by lipofection transfection with miR-294mimic to investigate the effect of miR-294on the proliferation of ESCs and expression of proto-oncogenes.Results:Baicalin suppressed the proliferation of ESCs, presenting as the longer population Td, obvious G2/M arrest and the reduced cell distribution in S phase. Baicalin downregulated the expression of proto-oncogenes c-jun and c-fos, as well as miR-294. Overexpression of miR-294shortened the Td of ESCs and rescued the prolonged Td induced by Baicalin, meanwhile it upregulated the expression of c-jun and c-fos and rescued the downregulation of these genes by Baicalin. The differences of the proliferation and expression level of proto-oncogenes and miRNAs of ESCs by Ginsenoside Rgl and Puerarin were not statistically significant.Conclusion:Baicalin suppressed the proliferation of ESCs by regulating the expression of proto-oncogenes c-fos and c-jun via miR-294.
