Part I:Expression of MIF in human colon carcinoma cell line LoVo and the significanceObject:To detect the expression of macrophage migration inhibitory factor (MIF) in human colon carcinoma cell line LoVo, and investigate the possible significance.Methods:Real-time quantitative PCR and Western blot were utilized to detect the expression levels of MIF mRNA and protein in LoVo and Fetal colon cell line (FHC), respectively.Results:Compared with FHC group, the expressions of MIF mRNA and protein in LoVo group both increased significantly.Conclusion:There is a possible relationship among the high expression of MIF in colon carcinoma cells and the development of colon carcinoma. Part II:Effects of MiR-451-mediated regulation of MIF on human colon carcinoma cell line LoVoObject:To investigate the effects of MiR-451-mediated regulation of macrophage migration inhibitory factor (MIF) on human colon carcinoma cell line LoVo.Methods:Lentivirus hsa-mir-451a was designed and used to infect LoVo cells to overexpress miR-451. Real-time quantitative PCR and Western blot were utilized to detect the expression levels of MIF mRNA and protein in LoVo and Fetal colon cell line (FHC), respectively. Proliferation of cells in each group was determined by MTT after infection.Results:After infected by lentivirus hsa-mir-451a, compared with un-infected group and control group, the expressions of MIF mRNA and protein in infected group both decreased significantly. And proliferation of cells in infected group was significantly inhibited.Conclusion:MIF is a potential miR-451gene target. Overexpression of miR-451can suppress the LoVo through direct suppression of MIF.
