Bidirectional promoters as a special kind of promoter are identified in diverse organisms with widely varied genome sizes, including bacteria, yeast, mammals, and plants. However,little research has been done on any individual endogenous bidirectional promoter from plants. In maize, in view of the great potential of bidirectional promoter in the application of multigene expression, so cloning and identification of seed specific bidirectional promoter and to study the gene regulation mechanism has important theoretical significance and application value. These characterized bidirectional promoters could serve as promoter resources for gene stacking and metabolic pathway engineering in maize and other monocot plant species. In addition, the seed specific bidirectional promoters with China's independent intellectual property rights can provide technical and material reserves for the long-term development of biotechnology industry in Maize Breeding in China.- Cloning and identification of a maize embryo specific bidirectional promoter PZmBD1 that can be applied in agricultural biotechnology. Moreover,we proposed a bidirectional promoter transcriptional regulatory model through the comprehensive analysis of bidirectional promoter sequences and the corresponding experimental verification.-On the basis of PzmBD1, endosperm-specific cis elements are fused to its sequence at specific location that forms eight chimeric promoters. Six are embryo specific bidirectional promoters with different promoter strength and the other two are seed specific promoters.-Establishing a maize gene multiple expression system that based on bidirectional promoter and 2A linker peptide, so as to provide method and technology support for the production of important bioactive protein or other secondary metabolites using maize seed as bioreactor and homologous recombination or heterologous reconstruction of complex metabolic pathway.
