Backgrouds:Objective: Cervical cancer is one of the most common malignant tumor of gynecologic.Recent studies have demonstrated that the abnormal expression of micro RNAs was involved in cervical cancer carcinogenesis and progression.To explore the expression of micro RNA-195 in cervical cancer.Discover the possible effect of miR-195 and its mechanism.Methods: qRT-PCR was used to detect the expression level of micro RNA-195 in cervical cancer tissues and cell lines.After transfection,MTT assay,cell migration and invasion assay,western blot and Dual-Luciferase report assay were performed in human cervical cancer cells.Resuls: Micro RNA-195 was down-regulated in cervical cancer tissues and cell lines.Additionally,up-regulation of micro RNA-195 and knockdown of HDGF was both inhibited cervical cancer proliferation,migration and invasion.Moreover,Dual-Luciferase report assay revealed that HDGF was a direct target gene of micro RNA-195.western blot showed that micro RNA-195 mimics inhibited HDGF protein level,whereas micro RNA-195 inhibitor enhanced HDGF expression of protein level.Conclusion: MicroRNA-195 targeted HDGF to inhibit tumorous behaviors of cervical cancer.These findings also suggested that micro RNA-195 could be a therapeutic biomarker of cervical cancer in the future.
