BackgroundPancreatic cancer is an aggressive disease with a particularly poor prognosis for a large percentage of cancer deaths.Gemcitabine is currently the first-line adjuvant chemotherapy for pancreatic cancer,but the survival rate of pancreatic cancer patients is still insufficient.In recent years,a number of clinical trials of gemcitabine and other drugs in the treatment of pancreatic cancer,but the effect is still not ideal.Therefore,it is important to find more effective drugs for pancreatic cancer,or drugs that combine gemcitabine with a better therapeutic effect.Baicalein is one of the main active ingredients of Scutellaria baicalensis Georgi.It is also the main constituent of scutellaria baicalensis.In recent year,a lot of studies have shown that baicalein has a variety of pharmacological effects,especially for a variety of tumors have a significant inhibitory effect.After a short dose of baicalein treatment,tumor cells can be effectively inhibited,and when a large dose of baicalein treatment for a long time,can induce apoptosis of tumor cells.Therefore,we considered whether small doses of baicalein combined with gemcitabine can effectively inhibit the growth of pancreatic cancer cells,as a potential treatment.Methods(1)Inhibitory effects of baicalein combined with gemcitabine in pancreatic cancer cells in vitro1.Inhibitory effect of baicalein,gemcitabine and two drugs on proliferation of pancreatic cancer cell line CFPAC-1 by MTT assay;2.Hoechst33258 staining,fluorescence microscope observation of baicalein and baicalein combined with gemcitabine on pancreatic cancer cells;3.Effect of baicalein and gemcitabine combined with two drugs on apoptosis of pancreatic cancer cells by Annexin-V APC/7-AAD double staining;4.Effects of baicalein,gemcitabine and two drugs on expression of apoptosis-related proteins in pancreatic cancer cells.(2)Baicalein combined with gemcitabine treatment of pancreatic cancer in vivoThe CFPAC-1 cells were resuspended in serum-free DMEM at a concentration of 1x107 cells/ml.A total volume of 0.2 ml cell suspension was then injected subcutaneously into the right anterior armpit of nude mice to establish a xenograft model.We start to inject saline,B AI,GEM and combination when the tumor growth to 2-4 mm.The volume of subcutaneous xenografts were recorded in nude mice for 28 days.And then euthanized nude mice,take the tumor for weighing and immunohistochemical staining.Results1.Baicalein or gemcitabine significantly inhibited the proliferation of human pancreatic cancer cell line CFPAC-1,and the drug concentration was in accordance with the tolerance,and the combination of two drugs on cell proliferation inhibition effect is more significant;2.Combination of baicalein and gemcitabine could induce the apoptosis of human pancreatic cancer cell line CFPAC-1;3.Baicalein combined with gemcitabine induced apoptosis of pancreatic cancer cells through regulating the expression of apoptosis proteins play a role;4.Baicalein combined with gemcitabine significantly inhibited the growth of human pancreatic cancer xenografts in nude mice,and the effect was better than that of the single drug.ConclusionsBaicalein can inhibit the proliferation of pancreatic cancer cells and induce differentiation.In vitro and in vivo experimental studies have shown that combination of baicalein and gemcitabine can significantly inhibit the proliferation of human pancreatic cancer cells,the effect is better than single drug treatment.Baicalein and gemcitabine combination will be the new way of treatment of pancreatic cancer.
