ObjectiveThrough feeding C57bl/6j mice with high fat diet,the present dissertation aims to observe the effects of JTXK and Yiqi Wenyang component on their weight,food intake,body fat,OGTT,blood glucose,blood lipid and adipose tissue thermogenic function.Meanwhile,with the help of testing White adipose tissue browning,the potential mechanism of JTXK and Yiqi Wenyang is explored.Methods8-week-old C57BL/6J male mice were fed with high-fat diet for 12 weeks.The standard is that they had ± 1.96 times of weight on the standard deviation or outweighted 20%of the standard model.These mice were randomly divided into 7 groups,which were control,normal JTXK,Rb1,Rg3,Cinnamaldehyde,pioglitazone respectively.In the following 8 weeks,all mice were administered.Food intake,body weight were recorded every week.After 8 weeks,the glucose tolerance and body fat detection was tested.After that,serum lipids and free fatty acid levels were detected.For general morphological observations,adipose tissues were stained by haematoxylin and eosin.Thermogenic function,glucose and lipid metabolism related gene mRNA expressions in adipose tissue were detected by real-time PCR.Immunoblotting was performed to detectUCP-1?PPAR??PGC-1?.PRDM16 protein expression in multiple adipose tissues.ResultsWeight and food intake:After the intervention of JTXK,the weight of mice was stable.From the 6th week of intervention,the weight gradually decreased,which was significantly lower than the control group(p<0.05).The food intake was lower than control group at 4th week(p<0.05).The result indicats that the impact of JTXK is related to time.Yiqi Wenyang group proved that the weight of mice was gradually decreased from the 6th week after the intervention,compared with the control group,the difference was statistically significant(p<0.05).The effect of Rbl on weight loss began to appear at 4th week after administration,and the difference was statistically significant(p<0.05).Rg3 had a significant weight loss at 5th week than the control group(p<0.05).The cinnamaldehyde group had a lower weight.An average reduction of 2.0g.Rb1 and Rg3 gained weight by 1.8g and 2.0g.There were significant difference in weight change between the three groups and the control group(p<0.05).The result suggested that Yiqi Wenyang component contributes to the gaining weight obviously,while Cinnamaldehyde,Rbl and Rg3 group showed reduction in food intake from the 5th week after administration,and the difference was statistically significant(p<0.05).Body fat test results:The body fat content of the mice in the model group was significantly higher than that of mice in the normal group,the body fat content was about 35%,and the body fat content of the mice in hypoglycemic Xiaoke granule group was 20.2%,which was significantly lower than that of the model group,the difference was statistically(P<0.05).The body fat content of mice in guinea saponin group,ginsenoside Rbl group and ginsenoside Rg3 group were 20.7%,23.8%and 22.3%respectively,which were significantly lower than those in model group(p<0.05).Therefore,Yiqi Wenyang component can reduce the total amount of fat for obese mice,and improve body composition to some extent.Blood lipids and free fatty acids:The levels of TC,TG and LDL-C in obese mice were significantly lower than those in model group(P<0.05).The levels of serum HDL-C were significantly lower in obese mice than in model group(p<0.05).Hypoglycemic granules and pioglitazone could significantly reduce the levels of FFA in obese mice,which were significantly lower than those in model group(p<0.05).After 8-weekdrug intervention,cinnamaldehyde showed a good effect on reducing the levels of TC,TG and LDL-C in obese mice and increasing the level of HDL-C in serum,which was significantly lower than that in model group(P<0.05),and the difference was statistically significant(p<0.05).Although the levels of TC and LDL-C in the two groups were lower than those in the model group,the levels of TC and LDL-C were significantly lower than those of the model group(P<0.05).The difference was not statistically significant(P>0.05).and the levels of FFA in obese mice were significantly lower than those in model group(p<0.05),and the difference was statistically significant(p<0.05).OGTT:After 8-week administration,the blood glucose showed no significant difference in fasting blood glucose between among the groups.After 30 minutes of glucose intragastric administration,the blood glucose of the model group began to increase significantly,and the hyperglycemic state lasted for 60 min and 120 min after oral glucose(P<0.01).The levels of blood glucose in pioglitazone and hypoglycemic Xiaoke granules were significantly lower than those in the control group(P<0.01),and the levels of blood glucose in the two groups were significantly lower than those in the normal mice(P<0.01),and the difference was statistically significant Blood glucose levels are close.AUC results showed that pioglitazone and hypoglycemic granules could significantly reduce the area under the curve of OGTT in obese mice,suggesting that 8-week administration could significantly improve the glucose tolerance of obese mice.(P<0.05),and the blood glucose level of normal mice was close to that of-normal mice.The blood glucose level of guanidine group,ginsenoside Rb1 group and ginsenoside Rg3 group was significantly lower than that of model group(P<0.05).AUC results showed that cinnamaldehyde,ginsenoside Rbl,ginsenoside Rg3 significantly reduced the area under the curve of OGTT in obese mice,which suggested that 8-week administration could significantly improve oral glucose tolerance in obese mice.HE:The volume of adipocytes in the white and brown adipose tissue of the hypoglycemic Decoction granule group and the pioglitazone group decreased,the content of the lipid droplets significantly decreased,and the multi-cavity beige fat cells increased.Cinnamaldehyde,ginsenoside Rbl group,ginsenoside Rg3 group of white and brown adipose tissue fat cells were smaller,significantly reduced lipid droplets,and can be seen multi-cavity beige fat cells increased,indicating that Yiqi Wenyang components to a certain extent.The result showed that Yiqi Wenyang components can improve white fat cell hypertrophy,and promote white fat cells brown.PCR and Western Blot:The expression of UCP-1,PGC-la and PPARy in white adipose tissue(WAT)of obese mice increased,amd the expression of UCP in white adipose tissue(WAT)-1,PGC-la and PPARy improved.The expression of UCP-1.PGC-1? and PPAR? in brown adipose tissue(BAT)of obese mice increased;expression of UCP-1,PGC-1? and PPARy in brown adipose tissue(BAT)of high-obese mice were enhanced.The expression of PPARy,PGC-1?,PRDM16 and UCP-1 in white adipose tissue(WAT)of obese mice can increase,and the expression of qi-yang yang-yang component in white adipose tissue(WAT)PPARy,PGC-1?,PRDM16 and UCP-1 in the brown adipose tissue(BAT)of obese mice increased,the expression of PPARy,PGC-la,PRDM16 and UCP-1 in brown adipose tissue(BAT)improved.The expression of PPAR?,PGC-1?,PRDM16 and UCP-1 in brown adipose tissue(BAT)of obese mice increased.Conclusions1 Hypoglycemic Decoction tablets weight loss lipid-lowering mechanism may be related to its promotion of white fat brown.2 The promotion of white fat brown causes may be related to Qi and Yang components.
