The improvement of antitumor therapeutics has been limited by several factors:(1)The primary challenge of antitumor therapeutic was the failure of most therapeutical agents to accumulate in the tumors,additionally causing serious systemic side effects;(2)the release of drug from vehicles,however,was not controlled and was relatively slow;(3)sole-therapeutics was not efficient to all types of tumors,and it might hardly realize totally ablation of tumors;(4)diagnostic method was necessary to monitor the location and size of tumor,as well as biodistribution of active agents during therapeutics.This study has employed gold nanorods(GNR)to assist nanocarriers to co-accumulate in tumors depending on enhanced permeability and retention(EPR)effect which realized passive targeting delivery;in addition,pH(low)insertion peptide(pHLIP)was used to decorate the vehicles,facilitating active targeting accumulation under acidic microenvironment of tumor site.Near infared(NIR)light,a non-invasive and permeable remotely-controlled trigger,was used combined with phothermal agents to provide heat,thus induce drug release from the photo-responsive nanocarriers after they specifically accumulated in tumors.The whole story has used photothermal agents combined with antitumor drug and photosensitizer loaded photothermal-triggered release nanocarriers to realize photothermal therapy(PTT)/chemotherapy and PTT/photodynamic therapy(PDT),respectively.In addition,this research has loaded imaging agents,such as fluorescent molecules,onto the photoresponsive nanocarriers,or employed imaging agents,such as gold materials,as nanocarriers to fabricate theranostic antitumor systems.In summarization,I have designed two theranostic systems to realize synergistic antitumor effect:(1)I have designed a tumor-targeting accumulated and locally triggered-release nanocarrier system based on GNR and thermosensitive liposomes(TSL).PTT,chemotherapy,and in vivo fluorescent imaging were simultaneously achieved under NIR light irradiation;(2)I have further improved a novel NIR light-induced triple-responsive antitumor therapeutic system based on targeting peptide functionalized hollow gold nanospheres(HAuNS),pHLIP,and photosensitizer,Chlorin e6(Ce6).This novel platform performed PTT-releasing-PDT step-by-step therapeutic process if NIR light was provided.
