Nanotechnology is a novel high-technology industry which shows a rapid evelopment in recent decades.The applications of nanomaterials have been extended to so many fileds,such as military,industry,medical and other related scientific fields.Zinc oxide(ZnO)is a kind of traditional detal material which has wide applications.Given the wide application of zinc oxide nanoparticles(ZnO NPs),the health hazards of these particles have attracted extensive worldwide attention.However,the information of the related toxicological mechanism remains obscure.Studies have showed that nanomaterials can penetrate into the brain through the brain blood barrier or olfactory brain pathways,and induce significant brain fucntion injuries.In the preliminary study of our group,we found that,after pregnant rats were exposed to ZnO NPs,the offsprings showed significant brain functions injuries which manifested as learning and memory ability impairment.This research has developed the microglials and neuro models via BV2 cells lines and PC 12 cell lines.Cells toxic effects after ZnO NPs treatment with various doses were detected,and the toxic differences between various doses were contrasted and the related mechanism were revealed.Further,the role of zinc ions which released from ZnO NPs in the toxicological mechanism were demonstrated.First chapter.The cytotoxic effects on the BV2 and PC12 cell lines after ZnO NPs treatment.We obtained the related materials informations via detecting the material characterization,and successly obtained the microglials and neuro models.Our study showed that ZnO NPs could induce various toxic effects after treatments with range of concentrations.Moreover,apoptosis and autophage were main toxic effects after ZnO NPs exposure,and oxidative stress played an important role in related toxicological mechanisms.Second chapter.The role of zinc ions released from ZnO NPs in related biophysical mechanismsZnO NPs showed different zinc release ability in different acidic environments with various doses.The acidic environment induced mass of zinc ions release and this effect played an important role in related toxicological mechanism.Third chapter.The cytotoxic effects on the BV2 and PC12 cell lines after ZnO NPs treatment.ZnO NPs with low doses promoted the cell proliferation via regulating the cell cycle,and the zinc ion release ability played a significant role here.Moreover,ZnO NPs pomoted the phosphorylation levels of ERK and Akt,which might be involved in promoting cell proliferation.Fourth chapter.The cytotoxic effects on the BV2 and PC12 cell lines after ZnO NPs treatment.ZnO NPs at toxic doses induced the cytoskeleton disruption,mitochondrial function impairment.
