| Purpose: This study focused the two key points of the happening and development of coronary atherosclerosis-starting with Arterial wall lipid deposition and abnormal cell proliferation,studying through the coronary heart Qinghua scattered in the prevention and treatment of atherosclerosis,and then exploring its anti-atherosclerosis mechanism of action for the “extravasted blood can turned into sputum,then turned into water”theory to provide reliable Experimental evidence.Method: 1.The establishment and evaluation of atherosclerosis rabbit model Sixteen normal breed male New Zealand rabbits were randomly divided into blank group(n = 8)and model group(n = 8).The blank group was fed with normal diet.The model group was given high fat diet and immunocompromised to prepare experimental atherosclerosis model.Continuous feeding for 10 weeks.Serum levels of total cholesterol(TC),triglyceride(TG),low density lipoprotein cholesterol(LDL-C),high density lipoprotein cholesterol(HDL-C).Observing the results of the whole oil red O staining of the aortic arch,and then determine whether the model is successful or not.2.The effect of Guanxinqinghua Powder on Blood Lipid and Plaque in Rabbits with Arteriosclerosis 60 healthy male New Zealand rabbits were randomly divided into blank group,model group,positive drug group,low dose group of traditional Chinese medicine,middle dose group and high dose group of Chinese medicine,and each group has 10 rabbits.Except the blank group,all the other groups were given high-fat diet + immunological damage to make experimental atherosclerosis model.At the same time,the low,middle and high dose groups of Chinese medicine were given 6.7g /kg/d,13.4g/kg/d and 26.8g/kg/d respectively.Positive drug group to simvastatin suspension 3mg/kg/d gavage,continuous gavage observation 12 weeks.Measuring the level of TC,TG,LDL-C and HDL-C.3.Study on the mechanism of Guanxinqinghua Powder against atherosclerosis Serum superoxide dismutase(SOD),malondialdehyde(MDA),malondialdehyde(MDA)and malondialdehyde(MDA)were measured by enzyme-linked immunosorbent assay(ELISA).Western blot was used to detect the reduced nicotinamide adenine dinucleotide phosphate oxidase 4(Nox4)(oxidase p22 phox protein,peroxisome proliferator-activated receptor γ(PPARγ),ATP-binding cassette transporter A1(NF-κB)were detected by Western blot.ABCA1),ATP-binding cassette transporter G1(ABCG1),B-type scavenger receptor(SR-B1),CD36 expression.The expression of Nox4,p22 phox,PPARγ,ABCA1,ABCG1,SR-B1 and caveolin-1 m RNA were detected by RT-PCR.4.Protective Effect of Guanxinqinghua Powder on Vascular Endothelial Cells Western blot was used to detect the the expression of basic Fibroblast Growth Factor,b FGF,extracellular regulated kinase1/2,ERK1/2,Fibroblast growth factor receptors 1,FGFR1)and p21 ras.Results: 1.The rabbit atherosclerosis model was successfully established by high fat diet and immunological injury.2.The levels of TC,TG and LDL-C in the atherosclerotic rabbits were significantly higher than those in the control group(P <0.05).The lipid-lowering effect was compared with simvastatin(P> 0.05).The positive drug group and the high,middle and low dose group saw the formation of lipid stripe,but the positive drug group and the middle and high dose group compared with the model group,the endometrial red patchy area was significantly reduced,and the model group of red Atherosclerotic plaques form a sharp contrast.3.Guanxinqinghua powder can increase the content of SOD,and decrease the content of MDA effectively,but middle dose is the most effcective and it can reduce the activity of Nox4 and p22 phox cells,and the levels of ocidative stress effectively.4.Coronary acid clearance inhibited the expression of CD36;increased ABCA1,ABGA1,SR-B1 and reduced the expression of PPARγ.5.Guanxinqinghua powder can reduce the expression of FGFR1,b FGF,p21 ras,ERK1/2 protein.Conclusion: 1.Guanxin Qinghua powder can significantly improve the blood lipid levels of atherosclerotic rabbits and reduce the formation of endometrial lipid striation and atherosclerotic plaque.2.One of the mechanisms of atherosclerosis may be anti-oxidative stress: Regulating the activity of Nox4 and p22 phox cell membrane NADPH oxidase,and reducing the level of arterial oxidative stress may be role that Guanxinqinghuasan plays in coronary heart and mechanism of antioxidant stress.3.The second Mechanism may be Inhibiting the expression of CD36,reducing the intake of macrophage cholesterol;increasing the expression of ABCA1,ABGA1,SR-B1 and caveolin-1 in the regulation of macrophage cholesterol balance,to strengthen the reverse transport of cholesterol,the mechanism of consideration and activation or increase the expression of PPARγ.4.Guanxinqinghua powder can inhibit Ras / Raf / MEK / ERK signaling pathway,thereby reducing cell proliferation and differentiation,play a role in protecting cells. |
