In today's highly mobile,interdependent and interconnected world,infectious diseases are a serious global problem,spread at a very fast pace and threat to public health.In combating infectious diseases,rapid and accurate diagnostic tests are required for pathogen identification except for developing new drugs.Aptamers are ssDNA or RNA oligonucleotides and screened by using a Systematic Evolution of Ligands by Exponential enrichment(SELEX)in vitro.Aptamers can specifically recognize their targets and regulate their functions.A number of nucleic acid aptamers have been identified and employed in many types of biosensing platforms,and also played a specific role in the study of the interaction between pathogen and host.The aptamers,targeting MDPV and AMDV for VP2 protein,have been identified and employed as affinity receptors in combination with unmodified gold nanoparticles and antibodies.And the two diagnosis sensors have good specificity and sensitivity compared to the classical methods.After 48h period post-transfection of ADVa012 aptamer in the AMDV infection system,47%lower virus concentration was observed at 4?M,whereas there was no inhibition by the random library.The half-life of ADVa012 aptamer was about 31.68h.The binding region of the ADVa012 may principally localize inside the P4 fragment(aa397-aa474)of VP2 protein.The above results showed that,the aptamer screening and application platform has been established targeting varous target,such as whole virus and purified protein.The aptamers have been screened and used to establish high sensitivity detection methods.And the selected ADVa012 aptamer have well inhibitory effect and long-term inhibition effect in vitro.
