The output and scale of kelp planting in China is the largest in the world.At present,we have formed industrial systems with alginate,mannitol and iodine as the main products.But in the above systems,more than 50% of the ingredients become solid wastes.Not only it wastes a lot of natural resources,but also brings a series of environmental pollution problems.Edible-medicinal fungi polysaccharides,which have many biological activities,are the main active substances in edible-medicinal fungi.They also have great research value and medicinal potentiality.In this paper,we took industrial production of kelp waste residue as culture medium,solid fermenting edible-medicinal fungi,producing fungal polysaccharides.Furthermore the solid fermentated kelp waste products were developed to a new type of fermented stichopus japonicus bait.In this paper,the growth rate and yield of polysaccharides of 10 kinds of edible-medicinal fungi strains solid-fermented on kelp waste were screened.The highest solid fermentation growth rate was of the strain I30,and the highest polysaccharide yield were of strain R24 and A47.And then,biological activities were screened for the activities of antihydroxyl activity,immune enhancement activity and tumor inhibition rate.The solid fermentation products of R24 and A47 showed excellent biological activity.In the preliminary experiment of stichopus japonicas feed,R24 and A47 also had good growth promoting effects on stichopus japonicas,with the macrophage enhanced activity of 19.92±0.16% and anti-hydroxyl activity EC50 of 7.44±0.51?g/m L of R24 hot water extracts product at the concentration of 1.0mg/m L,and the macrophage enhanced activity of 16.44±0.12% and anti-hydroxyl activity EC50 of 58.36±3.11?g/m L of A47 hot water extracts product at the concentration of 1.0mg/m L.The screened A47 and R24 strains were further processed single factor experiment on solid fermentation conditions on kelp waste.And then two regression general rotary model were designed,the five factors and four levels response surface optimization of solid fermentation conditions of R24 and A47 were investigated,the growth rate models of two strains were obtained respectively,and finally the optimum conditions for solid fermentation of kelp waste were obtained.The repeated verification tests under optimum conditions showed a high fitting rate between the model predictions and the test result s.Furthermore,the polysaccharides yield,tumor inhibition rate and immune enhance activity of solid fermentation products were tested.In the subsequent stichopus japonicus feeding experiment,in order to increase the specific growth rates,feeding rate and survival rate of stichopus japonicus,we optimized the stichopus japonicus bait recipe,and obtained optimal two recipes of HN15-7 and HN5-4.The results showed the new type of fermented stichopus japonicus bait,which was mainly composed of solid kelp waste fermentation products,could replace the finished feed of stichopus japonicus G4 in the preliminary experiment and promote the feeding,digestion and growth rate of stichopus japonicus.The survival rate of stichopus japonicus in each treatment group was measured under extreme experimental conditions.The HN5-4 significantly affected the survival rate of stichopus japonicus in extreme environment(P<0.05),which showed its effect on enhancing the immune activi ties of stichopus japonicus.Assay of digestive enzyme and immune relative enzyme activities showed enhancing effect of specific growth rate and disease resistance of stichopus japonicus of fermented kelp waste bait.Finally fungal polysaccharides of each product were added to its bait at different concentration.Results showed fungal polysaccharides from fermented kelp waste could enhance the amylase activity,the specific growth rate,phagocytosis rate and nonspecific immune enzyme(lysozyme,superoxide dismutase and acid phosphatase)activities of stichopus japonicas,which also showed promote growth and immune enhancement effects on stichopus japonicas.In summary,this paper studied the solid remaining waste problem in the kelp processing industry in coastal areas,proposed method for producing stichopus japonicus bait with fungus fermented on kelp waste,optimized the conditions for solid fermentation of kelp waste and analyzed biological function of fungal polysaccharides from the fermentation.Finally,the promote growth and immune enhancement effects on stichopus japonicas from fungal polysaccharide in fermented kelp waste bait were found.The experiment of environmental friendly stichopus japonicus bait with the main component of kelp waste fermentation products will provide itself the theoretical basis and technical support.Results of immune enhancing activities of stichopus japonicas bait provides another option besides antibiotics during disease outbreak.
