The thesis is mainly focused on studies of palladium-catalyzed C-H bond functionalization at C2 of indole in water and palladium-catalyzed N-H functionalization of indole in Water.It consists of the following three parts: Chapter 1.Application of C-H functionalization strategy to construct quaternary carbon center in synthesis.(review)The reaction of C-H bond direct functionalization has evolved into an effective tool for constructing C-C bonds and C-X bonds,and is increasingly being used in the synthesis of natural products and drug molecules;at the same time,in view of the important position of constructing quaternary carbon center in the synthesis,this chapter mainly reviews the application progress of constructing quaternary carbon center in the synthesis by using the C-H bond functionalization strategy.Chapter 2 "Two-in-one" Strategy C-H Bond Functionalization Reaction.The "two-in-one" C-H bond functionalization strategy was briefly introduced.A reaction of palladium-catalyzed C-H bond functionalization of indole at C2 position in water was developed by using this strategy.The reaction achieved cyclization of tryptamine derivatives with ketones and phenols respectively,to prepare a series of ?-tetrahydrocarbazole compounds,and an in-depth mechanism research was carried out.Chapter 3 "Dearomatization-Rearomatization" Strategy Indole N-H Bond Functionalization Reaction.A series of coupling reactions of phenyl developed by our group using the dearomatization-rearomatization strategy were introduced briefly.On this basis,a reaction of palladium-catalyzed N-H bond functionalization of indole in water was developed by this strategy.The reaction was carried out to achieve the reduction coupling of the N-1 position of indoles with ketones,to prepare a series of indole N-alkylation products,and an in-depth mechanism research was carried out.
