Soybean cyst nematode is one of the important diseases of soybean production,and it is the main content of this paper to control the disease of the soybean cystis.This paper systematically studied the action mechanism of Sneb545 induced soybean resistance to soybean cystis,and revealed the interaction between simple bacillus cereus,soybean and soybean cysts.Sneb545 by bacterium fermented liquid coating processing soybean research after soybean root nematodes in the change in the growth and development,the root of organization structure change,soy protein related course related gene changes,soybean roots endogenous hormone changes,soybean root transcription of omics,soybean root metabonomics analysis,validation,nematicides active metabolic substances,has been clear about the bacterium Sneb545 induction of soybean resistance to soybean cyst nematode mechanism.The main results are as follows:1.Clear the bacterium Sneb545 within the root induction of soybean resistance to soybean cyst nematode worms growth changes in the situation,found that the bacterium Sneb545 induction of soybean not only reduce the soybean cyst nematode infection rate at 50.8 %,more inhibits the growth and development of soybean cyst nematodes,which at the highest rate 36.8 %,finally have the effect of disease resistance.2.The change of the root tissue structure of the bacterium Sneb545 induced soybean resistance to soybean cystis nematode was clarified.Using paraffin section technology,in both infected soybean varieties in bacterium resistant soybean varieties Sneb545 processed soybean root phloem of wooden structure change,increase the lignin accumulation in the root,so as to improve the disease resistance of soybean.3.The changes of endogenous hormones induced by Sneb545 of bacterium were clarified.Bacterium Sneb545 treated root in auxin(IAA),gibberellin(GA),abscisic acid(ABA)and cytokinins(ZR),in different time points of the soybean resistance to nematodes content exist certain differences.4.It was clear that the bacterium Sneb545 induced the changes of related protein genes related to the resistance of soybean cystis.PR genes PR1,PR2,PR3 a,PR3b,PR5,PR9,PR10 and PR12 were selected to play a role in the process of soybean resistance.5.GC-MS technology was used for the first time to clarify the variation of the metabolites of the bacterium Sneb545 induced soybean resistance to soybean cystis.There were 15 metabolites associated with the anti-soybean cyst nematode.Clear bacterium Sneb545 induced three soybean resistance to soybean cyst nematode disease resistance mechanism:(1)The bacterium Sneb545 induction of soybean root nematode is one of the mechanisms of resistance by reducing the nematodes sources of nutrition,thus inhibiting nematodes in thesoybean root growth and development,to improve the ability of soybean disease resistance.(2)The bacterium Sneb545 can also induce the root of soybean to produce some active substances with nematodes to directly inhibit the growth and development of nematodes.(3)The bacterium Sneb545 induced soybean to produce disease-resistant mechanism similar to the disease-resistant varieties,thus improving soybean resistance to disease.6.RNA-seq high-throughput technology was used for the first time to clarify the differential gene expression of the bacterium Sneb545 induced soybean-resistant soybean cyst.In the 5 days of soybean inoculation nematode,there were 4185 up-regulated genes and 5298 down-regulated genes in the inoculated nematode worm and the unconnected nematodes.There were 2,833 up-regulated genes and 2,971 down-regulated genes in Sneb545 treatment of soybean grafts and unconnected nematodes.During the 10 days of inoculation of soybean nematodes,there were 3608 up-regulated genes and 3805 down-regulated genes in the inoculated nematode worm and unconnected nematodes.SSneb545 treatment of soybean grafts was 1059 and 997 genes were down-regulated.7.By metabolomics and transcriptome combined analysis,it was found that substances that may have toxic activity to cystic nematode were found,and the toxic effect of these substances on the second larva J2 of soybean cystic nematode was further verified by in vitro experiments.The metabolites of soybean root with nematode killing activity were methionine,piperine,palmitate and 4-vinyl phenol.
