Infectious bovine rhinotracheitis(IBR)is a highly contagious,infectious disease that is caused by bovine herpesvirus-1(BHV-1).In addition to causing respiratory disease,this virus can cause conjunctivitis,abortions,encephalitis and generalized systemic infections.The virus can cause immune suppression and secondary infection in cattle,seriously affecting the production performance of cattle.At present,the disease has been reported in major cattle-raising areas all over the world.China is also one of the major epidemic areas of the disease,which seriously endangers the development of cattle-raising industry in China.Jinshan District of Shanghai is a national demonstration area of modem agriculture.At present,there are 10 small and medium-sized dairy farms in the whole area,with a stock of nearly 10,000 cows.Infectious bovine rhinotracheitis has a certain degree of infection in Jinshan District,and pneumonia and abortion of dairy cows caused by infection often occur because of the frequent introduction of breeding cattle from abroad or other areas in China.However,there is no epidemiological data on infectious bovine rhinotracheitis in Jinshan District.Starting from the sero?epidemiological investigation,A method for quantitative PCR detection was established,forming a set of practical for integrated with Kingsoft the prevention and control of the project,to provide reference for scientific decision-making for local government.1.SERO EPIDEMIOLOGICAL SURVEY OF INFECTIOUS BRONCHITIS OF DAIRY COWS IN JINSHAN DISTRICT OF SHANGHAIIn this study,460 sera samples were collected:in 2016 from daily milks in 10 farms at Jinshan district,Shanghai.A serological investigation of infectious bovine rhinotracheitis was preformed using a commercial gE competition antibody detection ELISA kit.The results showed that the positive ratio is between 36.67%and 97.37%.It suggested the IBRV infection was popular and the strategies should be carried out to control the disease.2.ESTABLISHMENT AND APPLICATION OF QUANTITATIVE PCR FOR DETECTION OF GE GENE OF BOVINE HERPESVIRUS ITo accurately establish TaqMan probe real-time quantitative PCR(qPCR)method for Bovine herpesvirus(BHV-1),the primers and TaqMan probe specific to 1607-1704 bp of gE gene was designed according to the whole genomic sequence of BHV-1 representative strain.Using the viral DNA standard template,the stability,specificity,and sensitivity of the qPCR method were investigated.The results showed that,in the standard curve,R2 value was 0.999 with a high specificity.The sensitivity of the real-time PCR was less than 10 copies/?L.No cross reactions appeared to the other herpesviruses.The TaqMan probe qPCR method has the advantages to establish the BHV-1 detection method with high sensitivity and specificity.3.COMPREHENSIVE PREVENTION AND CONTROL OF INFECTIOUS BRONCHITIS IN DAIRY COWS IN JINSHAN DISTRICT OF SHANGHAIInfectious bovine rhinotracheitis(OBR)is a serious threat to the healthy development of dairy industry in Jinshan District,Shanghai,because the disease has the characteristics of latent infection,which had chances to be transmitted from the unhealthy cow when the transportation and detoxification of dairy cow,resulting in the risk of infection greatly increased.So,comprehensive strategies were carried out to the prevention and control of the disease,including diagnosis,detection,monitoring,immunization,disinfection,isolation,eulling,risk assessment and emergency management.It will be effective measures to control the disease.
